Özet

Amaç: Bu çalışmanın amacı, bazal tonusta elde edilen adrenerjik ve kolinerjik olmayan nöron kaynaklı gevşemelerde hiperpolarizan bir faktörün rolünün olup olmadığını ortaya koymaktı. Gereç ve yöntem: Çalışmada izole sıçan mide fundusu preparatı kullanıldı. Dokular fizyolojik koşullara uygun organ banyolarında uygun solüsyonlar içerisine alınarak çalışıldı. Bulgular: Mide fundusunun kısa süreli elektriksel alan stimülasyonu (0.1, 0.5, 1 Hz; 25 V; 1 ms; 10 saniye) ile ortaya çıkan cevaplar Nw-nitro-L-arjinin (100 MM ) ve lH-[l,2,4]oksalodiazolo[4,3-a]kinoksalin-l-one (10 MM) tarafından belirgin olarak azaltıldı. Deneylerde 1 MM'lık nifedipin ve 0.5 MM'lık verapamil dozları bazal tonusu belirgin derecede düşürdü. Kullanılan nifedipin konsantrasyonunda (0.5 MM) her üç frekansta da herhangi bir inhibisyona rastlanmadı. Sonuçlar: Sonuç olarak, sıçan mide fundusundaki nörojenik gevşemelerde nöron kaynaklı hiperpolarizan faktör veya benzeri bir hiperpolarizan faktörün rolü olduğuna dair bir kanıt elde edilememiştir.

Abstract

Objective: The aim of the present study was to investigate the possible role of a hyperpolarising factor in nonadrenergie noncholinegic nerve-mediated relaxations obtained in basal tone. Materials and Methods: Isolated rat gastric fundus preparations were used in the study. The tissues in organ baths were processed in convenient solutions matched with physiological environments. Results: The responses due to electrical field stimulation (0.1, 0.5, 1 Hz; 25 V; 1 ms; 10 sec) of the gastric fundus were greatly attenuated by Nw-nitro-L-arjinin (100 MM ) and lH-[l,2,4]oxalodiazolo[4,3-a]kinoxalin-l-one (10 MM). In the experiments, 1 MM and 0.5 MM concentrations of nifedipine and verapamil, respectively, decreased the basal tone of the tissue. There was no inhibition by nifedipine (0.5 MM) on the responses to electrical stimulation. Conclusions: As a result, there is no proof for a role of a nerve-derived hyperpolarising factor or of another similar hyperpolarising factor with respect to the neurogenic responses in the rat gastric fundus.
