Özet

Bu çalışma, tıbbi bitki Limnophila aromatica (Lamk.) Merr.'nın dokukültürü teknikleriyle hızlı ve verimli bir şekilde üretilmesi üzerineZeatin (ZEA) ve oksin kombinasyonlarının etkilerini sunmaktadır. L.aromatica'nın yaprak eksplantları 0.10-1.60 mg L-1 ZEA ve 0.10 mg L1 indol-3-asetik asit (IAA), indol-3-butirik asit (IBA) ve naftalen asetikasit (NAA) eklenmiş Murashige ve Skoog (MS) besin ortamında altıhafta boyunca kültüre alınmıştır. Hormon uygulamalarıkıyaslandığında eksplant başına maksimum sürgün sayısı 0.10 mg L1 ZEA + 0.10 mg L-1 IBA içeren MS besin ortamında elde edilmiştir(25.29 adet), ardından 0.10 mg L-1 ZEA + 0.10 mg L-1 IBA içeren MSbesin ortamında tespit edilmiştir (23.72 adet). Sürgün uzunluklarıZEA+IAA uygulamalarında 1.24-1.41 cm, ZEA+IBA uygulamalarında1.28-1.47 cm ve ZEA+NAA uygulamalarında 1.16-1.34 cm olarak eldeedilmiştir. En uzun sürgünler ZEA+IBA’nın kullanıldığı kültürortamlarında kaydedilmiştir. Rejenere sürgünler, 0.25 mg L-1 IBAiçeren MS ortamında köklendirilmiş ve köklenmiş bitkiler akvaryumabaşarıyla alıştırılmıştır. Sonuç olarak in vitro üretim için en uygunhormon kombinasyonu 0.10 mg L-1 ZEA + 0.10 mg L-1 IBA olaraktespit edilmiştir. Bu üretim protokolü, ticari olarak büyük ölçekteüretim için faydalı olabilir.

Abstract

This study presents the effects of Zeatin (ZEA) and auxin combinations on the fast and efficient production of medicinal plant Limnophila aromatica (Lamk.) Merr. by tissue culture techniques. The leaf explants of L. aromatica were cultured on Murashige and Skoog (MS) nutrient medium supplemented with 0.10-1.60 mg L-1 ZEA and 0.10 mg L-1 indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and naphthaleneacetic acid (NAA) for six weeks. The maximum number of shoots per explant compared to hormone applications were obtained in MS nutrient medium containing 0.10 mg L-1 ZEA + 0.10 mg L-1 IBA (25.29), followed by in the MS nutrient medium containing 0.10 mg L-1 ZEA + 0.10 mg L-1 IBA (23.72). The shoot lengths were obtained as 1.24-1.41 cm in ZEA + IAA applications, 1.28-1.47 cm in ZEA + IBA applications and 1.16-1.34 cm in ZEA + NAA applications. The longest shoots were recorded in culture media using ZEA + IBA. The regenerated shoots were rooted on MS medium containing 0.25 mg L-1 IBA and then the rooted plantlets were successfully acclimatized in an aquarium. As a result, the most suitable hormone combination for in vitro production was determined as 0.10 mg L-1 ZEA + 0.10 mg L-1 IBA. This production protocol may be useful for commercially large scale production.
