Özet
Bu çalışmada, çeşitli hayvan türlerine ait etlerin ve bu etlere yapılan katkı seviyelerinin belirlenmesi amacıyla, bazı et proteinlerinin ayırımında Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis (SDS-PAGE) tekniğinin kullanılabilme imkanları araştırılmıştır. Et türlerinin ayırımında spesifıc protein bantları kullanıldı. Araştırmada çiğ sığır, domuz, koyun ve at etleri ayrı ayrı ve ağırlıklarına göre muhtelif seviyelerde ikili (0/100, 5/95, 25/75, 50/50, 75/25, 95/5, 100/0) karışımlar şeklinde hazırlanmıştır. Hazırlanan örneklerdeki tür çeşidi ve bunların yüzde nispetleri SDS-PAGE tekniği ile protein profilleri yine molekül özettikleri bilinen standart protein markerleri ile karşılaştırılarak, teşhis ve tespitler gerçekleştirilmiştir. SDS-PAGE tekniğinin, et karışımlarındaki türlerin ve bunların karışımlardaki oranlarının % 5-10'a kadar tespitinde başarı ile kullanılabileceği ortaya konmuştur.
Abstract
In this study, for evaluating the effectiveness of Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis (SDS-PAGE) to identify the animal species in raw meat species adulteration in binary mixture was invastigated. Characteristic banding patterns of proteins for each species were used in identifying the existence of other species in a meat mixture. Electrophoretic patterns of water soluble proteins from single meat species and binary mixtures (0/100, 5/95, 25/75,50/50 75/25, 95/5, 100/0) by weight were presented. In this study, raw beef, pork, sheep and horse meat was individually prepared and mixed in various ratios based on flesh weight and then the protein patterns were compared with standart proteins markers. The species specific band decreases with decreasing the weight of flesh used in the binary mixture, and at 5% mixture, this band present in mixed samples. SDS-PAGE was an effective method to differentiate and identify species of raw meat in the binary mixture sample. Meat species adulteration at concentration as low as 5-10 % by -weight could be detected without risk.
