Trabzon (Karadeniz) Deniz Sedimentlerinden Elde Edilen Denizel Aktinomisetlerin Sekonder Metabolit Biyosentez Genlerinin Taranması

Özet
Bu çalışmada, Trabzon (Karadeniz) deniz sedimentlerinden elde edilen aktinomiset izolatlarının aktif sekonder metabolit üretme kapasitesi moleküler tekniklerle araştırılmıştır. Toplamda 24 aktinomiset, sekonder metabolit biyosentez genleri PKS/NRPS varlığı bakımından PCR tabanlı incelenmiştir. Yapılan PCR sonuçlarına göre Trabzon deniz sedimentlerinden elde edilen aktinomisetlerin %25'i PKS-NRPS gen bölgelerini birlikte, %58'i ise sadece NRPS gen bölgelerini bulundurduğu belirlenmiştir. PCR verileri değerlendirildiğinde izolatların ribozomal olmayan yolla peptid yapıda aktif sekonder metabolit üretim kapasitesinin PKS yolağı ile sekonder metabolit üretme kapasitesinden yüksek olduğu bulunmuştur. Ayrıca aktif sekonder metabolit üretimi için Karadeniz deniz sedimentlerinin yüksek potansiyel barındırdığı belirlenmiştir.
Abstract
In this study, active secondary metabolite production capacity of actinomycete isolates obtained from Trabzon (Black Sea) sea sediments was investigated by molecular techniques. Totaly 24 actinomycetes were investigated by PCR based on the presence of secondary metabolite biosynthesis genes PKS / NRPS. According to the PCR results, 25 and 58% of actinomycetes obtained from Trabzon sea sediments were found to contain PKS-NRPS and only NRPS gene regions, respectively. When PCR data were evaluated, it was found that the production of the peptide form active secondary metabolite of the isolates by non-ribosomal way was higher than that of the secondary metabolite production by the PKS pathway. In addition, it has been determined that Black Sea marine sediments have high potential for active secondary metabolite production.
