Tavşan trekea düz kasında Beta-adrenerjik reseptör agonistleri ve fosfodiesteraz inhibitörlerinin gevşetici etkilerinde K+-kanallarının rolü

Özet
Tavşan trakea düz kasında yapılan bu in vitro çalışmada izoprenalin, salbutamol, teofilin ve amino filin ile alınan gevşeme cevapları üzerine K + kanal blokörleri tetraetilamonyum (TEA) ve glibenklamidin inhibitor etkileri araştırılmıştır. Karbakol (10 -6 M) uygulanarak kasılan dokularda, kümülatif tarzda ilave edilen izoprenalin (10 -9 -10 -4 M), salbutamol (3x10 -10 -3 M), teofilin (10 -9 -10 -4 M) ve aminofilin (10 -9 -10 -4 M) doza bağımlı tarzda gevşeme cevapları oluşturmuş ve elde edilen % maksimum gevşeme değerleri sırasıyla % 87.83 ± 2.89, % 75.83 ± 2.04, % 73.33 ± 2.55, % 69.83 ± 2.36 olarak bulunmuştur. Belirtilen ajanlara bağlı gevşeme cevaplarının K + kanal blokörleri TEA (10 -4 M) ve glibenklamid (10 -6 M) ile anlamlı olarak inhibe edildiği, izoprenalin ve salbutamol cevaplarının inhibisyon oranının daha yüksek olduğu görülmüştür. Kontrol EC 50 değerleri karşılaştırıldığında gevşetici etki güçlerinin izoprenalin > aminofilin > teofilin > salbutamol şeklinde olduğu saptanmış ve K + kanal blokörleri bu ajanlar için bulunan doz cevap eğrilerini anlamlı olarak sağa kaydırmıştır. Sonuç olarak izole tavşan trakea düz kasında yapılan bu in vitro çalışmada betaadrenerjik reseptör agonistleri izoprenalin ve salbutamolun bronkodilatör etkilerinin önemli ölçüde K + kanallarının aktivasyonu ile meydana geldiği, fosfodiesteraz inhibitörü olan teofilin ve aminofilinin bronkodilatör etkilerinde K + kanallarının daha az rol oynadığı saptanmıştır.
Abstract
The aim of this in vitro study was to investigate the inhibitory effects of K<sup>+</sup> channel blockers, tetraethylammonium (TEA) and glibenclamide, on the isoprenaline-, salbutamol-, theophylline- and aminophylline- induced relaxation in the rabbit trachea. In the tracheal strips pre-contracted with carbachol (10<sup>-6</sup> M), isoprenaline (10<sup>-9</sup>-10<sup>-4</sup> M), salbutamol (3x10<sup<-7</sup>-10<sup>-3</sup> M), theophylline (<sup>-9</sup>-10<sup>-4</sup> M) and aminophylline (10<sup>-9</sup>-10<sup>-4</sup> M) each produced concentration- dependent relaxation. Mean maximal relaxant responses to isoprenaline, salbutamol, theophylline and aminophylline in the tracheal strips were 87.83 ± 2.89 %, 75.83 ± 2.04 %, 73.33 ±2.55% and 69.83 ± 2.36 %, respectively. Relaxant responses of these agents were significantly inhibited by K<sup>+</sup> channel blockers, TEA (10<sup>-4</sup> M) and glibenclamide (10<sup>-6</sup> M). The inhibitory effects of K<sup>+</sup> channel blockers on teophylline- and aminophylline-induced relaxations were lower than those of isoprenaline and salbutamol. The rank order of potency of these compounds was isoprenaline > aminophylline > teophylline > salbutamol and K<sup>+</sup> channel blockers caused a displacement to the right of the dose response curves to these agents. Results indicate that the K<sup>+</sup> channels play an important role in the relaxant effects of β-adrenoceptor agonists meanwhile their role in the teophylline- and aminophylline- induced relaxation is less.
