Özet

Amaç: Özel Çalışma Modülü (ÖÇM) programı, Dokuz Eylül Üniversitesi Tıp Fakültesi'nin ilk üç yılına entegre edilmiştir. Laboratuvar araştırması kategorisinde olan ÖÇM çalışmamıza, 2017-2018 eğitim-öğretim yılında Dokuz Eylül Üniversitesi Tıp Fakültesi Dönem 2 düzeyinde dört öğrenci katılmıştır. Bu ÖÇM ile öğrencilere bağımsız öğrenme, bilimsel metodolojinin temel ilkeleri ve bilimsel araştırma sonuçlarının yazılı ve sözlü sunum yapma becerilerinin kazandırılması yanısıra sisplatin ile indüklenmiş testis hasarında alfa lipoik asitin p38 MAPK sinyal ileti yolağı üzerine olası etkisinin araştırılması amaçlanmıştır. Gereç ve Yöntem: ÖÇM’de Wistar albino sıçanlar, kontrol (n=7), sisplatin (16 mg/kg, intaperitoneal, i.p) (hasar grubu) (n=7) ve sisplatin+alfa lipoik asit (LA) (100 mg/kg, i.p) (tedavi grubu) grubu olmak üzere üç gruba ayrıldı. Testis dokularında total p38 MAPK ve aktive olan fosforile p38 (p-p38) sinyal ileti yolağı protein ifadesi western blot yöntemi ile, hücresel lokalizasyonu ise immunohistokimya ile değerlendirildi. ÖÇM ile ilgili deney analizleri bittikten sonra, öğrenciler yazılı bir rapor hazırladı ve sonuçlarını eğitim-öğretim yılının sonunda sözlü olarak sundu. Dönem sonu verdikleri geri bildirim ile de ÖÇM sürecini değerlendirdiler. Bulgular: ÖÇM sonunda, öğrenciler literatür tarama, bilimsel araştırma planlama, etik kurul başvurusu yapma, temel laboratuvar teknikleri ve analiz sonuçlarını değerlendirme ile ilgili deneyim kazandılar. Ayrıca, öğrenciler bilimsel makaleleri okumayı ve tartışmayı öğrendiklerini, laboratuvardan ve edindikleri araştırma becerilerinden memnun olduklarını belirttiler. Deneysel bulgular değerlendirildiğinde ise, sisplatin ile indüklenen hasar grubunda total p38 ve p-p38 protein ifadelerinin kontrol grubuna göre anlamlı olarak arttığı saptandı (p<0.05). Bununla birlikte LA ile bu artış anlamlı olarak azaldı (p<0.05). Immünohistokimyasal bulgularda da seminifer tübüllerde saptanan dağılım western blot bulguları ile benzerdi. Sonuç: Öğrencilerin bağımsız öğrenme becerilerinin geliştirilmesi ve bilimsel metodolojinin temel ilkelerini öğrenmeleri hedeflerinden oluşan bu ÖÇM, Tıp Fakültesi müfredatında yeni ÖÇM’lerin yapılandırılmasında yol gösterici olacaktır.

Abstract

Objective: The Special Study Modules (SSM) have been integrated into Dokuz Eylül University Medical School's educational activities for the first three years. In 2017-2018 academic year, four second-year students of Dokuz Eylül University Medical School participated in our SSM study, which was in the laboratory research category. The aim of this study was to investigate the possible effect of alpha lipoic acid on p38 MAPK signalling pathway in cisplatin-induced testicular damage, as well as to provide students with the skills of independent learning, knowledge on the basic principles of scientific methodology and with the ability to present scientific research results both orally and in writing. Material and Method: In SSM, Wistar albino rats were divided into three groups: Control group (n=7), cisplatin (16 mg/kg, intraperitoneal, ip) (damage group) (n=7) and cisplatin+alpha lipoic acid (LA) (100 mg/kg, ip) (treatment group). Total p38 MAPK and phosphorylated p38 (p-p38) signalling pathway protein expression in testicular tissues were evaluated by western blot method and cellular localization was assessed through immunohistochemistry. After the experimental analyses, the students prepared a written report and presented the outcomes orally at the end of the academic year. They assessed the SSM process through the feedback they gave at the end of the term. Findings: At the end of the special study, students gained experience in the literature review, scientific research planning, applying to an ethics committee, basic laboratory techniques and evaluating the findings of the analysis. Besides, the students stated that they learned to read and discuss scientific articles and were satisfied with the laboratory and the research skills they acquired. When the experimental findings were evaluated, it was ascertained that total p38 and p-p38 protein expressions in the cisplatin-induced damage group increased significantly compared to those in the control group (p <0.05). However, this increase lessened significantly when LA was administered (p <0.05). The immunohistochemical findings were similar to western blot results in terms of the distribution detected within the seminiferous tubules. Conclusion: This SSM, aiming at improving independent learning skills of the students and enabling them to learn the basic principles of scientific methodology, will be a guide for the formation of new SSMs within the curriculum of the Medical School.
