Özet
Bu çalışmada klinik örneklerden Mycobacterium tuberculosis saptanmasında kullanılan Bactec radyometrik yöntemi, M.tuberculosis'in tekrarlayan dizilerinden oluşturulmuş oligonükleotidlerin kullanıldığı polimeraz zincirleme reaksiyonu (PCR) ve M.tuberculosis rRNA'sının çoğaltılmasının "hibridization protection assay" ile birleştirildiği çoğaltılmış M.tuberculosis direk testinin (AMTD) karşılaştırılması amaçlanmıştır. Çalışmada toplam 100 klinik örnek üç yöntemle incelenmiştir. Bactec yöntemi ile PCR, Bactec yöntemi ile AMTD ve PCR ile AMTD yöntemleri arasındaki uyum oranları sırasıyla %88, %86 ve %88 olarak bulunmuştur. Klinik örneklerden M.tuberculosis saptanmasında üç yöntem arasında istatistiksel olarak anlamlı bir fark olmadığı görülmüştür. Duyarlılıkları Bactec kültür yöntemine benzer bulunan PCR ve AMTD yöntemlerinin, M.tuberculosis enfeksiyonlarının hızlı tanısında yararlı nükleik asit çoğaltma yöntemleri olduğu sonucuna varılmıştır.
Abstract
The aim of this study was to compare the three methods, Bactec radiometric method, polymerase chain reaction (PCR) by using oljgonucleotides based on the repetitive sequence of Mycobacterium tuberculosis and amplified M.tuberculosis direct test (AMTD, a test that combines M.tuberculosis rRNA amplification with the hybridization protection assay) used for the detection of M. tuberculosis from clinical specimens. A total of 100 clinical specimens were studied by these methods, and the agreements between Bactec method and PCR, Bactec method and AMID, and PCR and AMTD were found as 88%, 86% and 88% respectively. The statistical analyses showed that there were no significant differences between the three methods in the detection of M.tuberculosis from clinical specimens. Although the sensitivities of PCR and AMTD appeared to be similar to that of culture with Bactec system, the two methods based on nucleic acid amplification may be very useful for rapid detection of M. tuberculosis infections.
