Özet
Pseudomonas aeruginosa ile oluşan kronik akciğer enfeksiyonu, kistik fibrozisli hastalarda akciğer fonksiyonunun kaybından birinci derecede sorumludur. Bu çalışmada, kistik fibrozisli hastalardan izole edilen P.aeruginosa suşlarının arbitrarily primed-PCR (AP-PCR) yöntemi ile tiplendirilmesi ve böylelikle bir hastaya ait izolatların moleküler epidemiyolojik izleminin sağlanması amaçlanmıştır. İzolatlar iki rastgele primer kullanılarak AP-PCR ile incelenmiştir. Tüm izolatların antimikrobiyal duyarlılık testleri disk difüzyon yöntemi ile yapılmıştır. AP-PCR DNA amplifikasyon ürünleri incelendiğinde tüm hastalardan kronik P.aeruginosa kolonizasyonu süresince elde edilen seri izolatların her iki primer ile de aynı DNA paternine sahip olduğu görülmüştür. Bir hastaya ait iki izolatta ise ek bir DNA paterni saptanmıştır. AP-PCR tiplendirme yöntemi, koloni morfotipi ve antibiyotik duyarlılık paternleri benzer olmayan suşların aynı, özgün genotipte olabileceğini göstermiştir.
Abstract
Chronic lung infection with Pseudomonas aeruginosa is primarily responsible for pulmonary deterioration in cystic fibrosis (CF) patients. The purpose of this study was to type the P.aeruginosa isolates collected sequentially from CF patients, by arbitrarily primed-PCR (AP-PCR) to determine whether a single strain colonized the patients or there was co-infection with different strains during their follow-up. The isolates were analyzed by AP-PCR by using two arbitrary primers. Antimicrobial susceptibility testing of all isolates was performed by disc diffusion method. The AP-PCR DNA amplification products obtained with both of the primers revealed that in all of the patients, the serial isolates had the same DNA patterns during chronic colonization. However, an additional AP-PCR pattern was observed in two of the isolates of a patient. The AP-PCR typing also demonstrated that strains dissimilar in colony morphotype and of different antibiotic susceptibility patterns, could be of the same, unique genotype.
