Silikon yağının Staphylococcus aureus tedavisinde kullanılan antibiyotiklerle etkileşimi

Özet
Amaç: Staphylococcus aureus enfeksiyonları tedavisinde kullanılan antibiyotiklerin antibakteriyel etkinliğine silikon yağının etkisi in vitro olarak araştırıldı. Gereç ve Yöntem: Bu deneysel çalışmada, antibiyotik duyarlılık testleri National Commitee far Clinical Laboratory Standards 'in microbroth sulandırma yöntemine uygun olarak hazırlandı. Mueller Hinton (MH) sıvı besiyeri, test ortamı idi. Standart ATCC 29213 Staphylococcus aureus susu çalışıldı. Test edilen antibiyotikler amikasin, vankomisin ve ofloksasin idi. Çift olarak hazırlanan kuyucuklardaki antibiyotik dilüsyonlarının üzerine, birincilere silikon yağı (silikon grubu), ikincilere ise MH sıvı besiyeri (kontrol grubu) ve her ikisine de Staphylococcus aureus eklendi. Plaklar 18 saat 37°C'de etüvde inkübe edildi. Antibiyotiklerin minimal bakterisidal konsantrasyonunu (MBK) saptamak için, MH agar besiyerine subkültürler yapıldı ve koloniler 37°C'de 18 saat inkübasyondan sonra sayıldı. Bulgular: Antibiyotiklerin minimal inhibitor konsantrasyonları, silikon grubunda kontrol grubu ile aynı olduğu gözlendi. Silikon grubunda, ofloksasinin MBK'si kontrol grubu ile aynı bulunurken, vanko-misin ve amikasinin MBK'leri silikon grubunda kontrol grubuna oranla bir dilüsyon düşük bulundu. Sonuç: Staphylococcus aureus infeksiyonlarmda kullanılan antibiyotiklerden vankomisin, arnikasın ve ofloksasinin in vitro antibakteriyel etkinliğine, silikon yağının anlamlı bir etkisinin olmadığı görüldü. Bu çalışma in vitro olarak yapıldığından, bu sonuçlan in vivo teyit etmek için deneysel hayvan çalışmaları gerekmektedir.
Abstract
Purpose: To evaluate the interaction of silicone oil with the antibacterial activity of antibiotics against staphylococcus aureus. Materials and Methods: In this experimental study, antibiotic susceptibility tests were prepared according to the National Committee for Clinical Laboratory Standards microbroth dilution method. Mueller Hinton (MH) Broth was the test media. Standard ATCC 29213 staphylococcus aureus strain was studied. Amikacin, vancomycin and ofloxacin were tested for staphylococcus aureus. Two sets of antibiotic dilutions were prepared identically in the microplates. Silicone oil was put into one set of microplates (silicone group) and MH broth was put into the other set of microplates (control group) and staphylococcus aureus was then added into both sets of microplates. Trays were incubated at 37"C for 18 hours before reading. To determine the minimal bactericidal concentrations (MBC) of the antibiotics, subcultures were made onto Mueller Hinton agar, and the colonies were counted after 18 hours of incubation at 37°C. Results: The minimal inhibitory concentrations of the antibiotics in the silicone group were observed to be identical with the control group. The MBC of ofloxacin in the silicone group was found to be identical with the control group; whereas, the MBCs ofamikacin and vancomycin were found to be one-dilution lower in the silicone group, when compared with the control group. Conclusion: Silicone oil showed no significant effect on the antibacterial activity of vancomycin, amikacin and ofloxacin in vitro against staphylococcus aureus. As this is an in vitro study, experimental animal studies are required to confirm these results in vivo.
