Özet
Bu çalışmada bazı yerel anestezik (lidokain, bupivakain, prilokain) ilaçların ratlarda mesane üzerine in vitro etkileri araştırıldı. Denemeler izole organ banyosu ortamında gerçekleştirildi. Lidokain, bupivakain ve prilokain'in 6 farklı dozu denendi. Lidokainin 2. dozu (0.6 $\mu$g/ml) hariç diğer bütün dozları (1. doz: 0.2 $\mu$g/ml. 3. doz: 2 $\mu$g/ml. 4. doz: 6 $\mu$g/ml. -5. doz: 20 $\mu$g/ml, 6. doz: 60 $\mu$g/ml) mesane kaslarında gevşemeye neden oldu. Lidokainin 3. dozunun (p 0.05). Bu ilacın 4. (10 $\mu$g/ml) dozu mesane kasları üzerinde kasılma ve gevşeme bakımından herhangi bir değişikliğe neden olmadı, istatistiksel olarak prilokainin sadece 6. dozunun oluşturduğu gevşetici etki önemli bulundu (p<0.01).
Abstract
In this study, in vitro effects of liclocaine, bupivacaine and prilocaine on the urinary bladder muscle in rats were examined. The experiments were performed in isoloated organ-bath. Six different doses were tried for each local anesthetic. All doses of lidocaine (1. doz: 0.2 ug/ml. 3. doz: 2 $\mu$g/ml. 4. doz: 6 $\mu$g/ml, 5. doz: 20 $\mu$g/ml, 6. doz 60 $\mu$g/ml) except 2nd dose (0.6 $\mu$g/ml) caused relaxation on bladder muscles. The relaxation effects of the third (p<0.05), 4th and 6th (p<0.01) doses of the lidocaine were significant. It was observed that first (0.25 $\mu$g/ml), 5th (15 $\mu$g/ml) 6th (30 $\mu$g/ml) doses of bupivacaine showed relaxation, whereas 2nd (0.5 $\mu$g/ml), 3rd (1.5 $\mu$g/ml) and 4th (5 $\mu$g/ml) doses of this drug caused contractions on urinary bladder muscles. While first (0.5 $\mu$g/ml), 5th (30 $\mu$g/ml) and 6th (60 $\mu$g/ml) doses of prilocaine showed relaxation, 2rd (1 $\mu$g/ml) and 3rd (3 $\mu$g/ml) doses caused contractions. The 4th closes of this drug showed no sings of either relaxation or contraction. Only 6th doses of the prilocaine showed statistically significant relaxation (p<0.01).
