Köpeklerde erişkin Taenia hydatigena antijenine karşı antijenik spesifitenin SDS-PAGE ve western blot yöntemiyle araştırılması

Köpek, kurt ve diğer yabani karnivorlarm ince bağırsaklarında paraziti enen ve oldukça kozmopolit bir yayılışa salıip olan Taenia hydatigena'mn karnivorlarda teşhisi oldukça zor olup, yumurtaları Taeniidae ailesine bağlı diğer türlerin yumurtalanyla karışmaktadır. Bu çalışmada deneysel yolla enfekte edilen ile kontrol grubu köpek yavrularından elde edilen serumlar ve olgun T.hydatigena halkalarından hazırlanan antijen kullanılmıştır. Antijendeki proteinlerin molekül büyüklüklerinin belirlenmesi için antijenin SDS-PAGE yöntemiyle elektroforezi yapılmıştır. SDS-PAGE yöntemi ile separe edilen proteinler Western blot metoduyla nitfoselüloz membranlara transfer edilip, köpek serumları (deneysel enfekte ve kontrol grubu) ve ayrıca hazır ticari non-enfekte köpek serumu test edilmiş ve membranlarda açığa çıkan protein bantlanndan hangilerinin deneme grubu hayvanlar için spesifik olduğunu belirlemek için elde edilen bulgular diğer bantlarla karşılaştınlmıştır. Bu bulgulara göre Western blot yönteminde köpeklerde T.hydatigena'nın tanısında kullanılabilecek spesifik bantların molekül ağırlıklarının 24 ve 84 kDa olduğu tespit edilmiştir.
Abstract
Taenia hydatigena occurs in the small intestine of dogs, wolves and other wild carnivores and it is cosmopolitan in distribution. The diagnosis of this cestode is almost impossible via fecal examination in carnivores. Because the egg of this parasite is very similar to other species of Taeniidae family, which causes confusion in diagnosis. In this study the sera were obtained from the experimentally infected dogs and Control group dogs. Antigen was prepared from the T.hydatigena proglottids. The antigen was studied with SDS-PAGE to determine protein bands. These proteins were transferred to nitrocellulose membranes by the method of Western blotting. The dog sera and also commercial non-infected dog serum were tested on these nitrocellulose membranes. After that the results from the sera of experimental group were compared to the results of sera from Control group and non-infected dog serum to determine specific bands for this parasite. According to the results, the bands of 24 and 84 kDa are determined to be specific proteins in the diagnosis of T.hydatigena in dogs.
