Gebelik süsesince çinko verilen tuj ırkı koyunlarda serum lösin aminopeptidaz aktivitesinin belirlenmesi

Özet
Lösin aminopeptidaz (LAP) aktif bölgesinde çinko atomuna sahiptir ve gebelikte önemli rolleri bulunmaktadır. Bu çalışmada Tuj ırkı koyunlara gebelikleri süresince dışardan ilave olarak verilen çinkonun, gebeliğin serum LAP aktivitesindeki değişimlere etkisi araştırmak amaçlanmıştır. Çalışmada materyal olarak 3 yaşlı 20 adet Tuj ırkı koyun kullanıldı. Hayvanlar kontrol (n=10) ve deneme (n=10) olmak üzere iki gruba ayrıldı. Deneme grubundaki hayvanlara kontrol grubuna ek olarak 30 mg/kg çinko verildi. Gebelik boyunca ve gebelikten sonra hayvanlardan kan numuneleri alınarak, 15 dakika 3000 rpm’ de santrifügasyona tabii tutuldu. Gebelik (5 ay süresince) ve doğumdan sonra alınan serum numunelerinde LAP aktivitesi ölçüldü. Çalışmada deneme grubundan elde edilen LAP değerleri kontrol grubu ile karşılaştırıldığında istatistiksel olarak bir fark bulunamadı. Aylara göre LAP aktivitesinde ise en yüksek değerler gebeliğin son üç ayında tespit edildi. Her iki grupta da gebeliğin üçüncü ayından doğuma kadar istatistiksel olarak p<0.001 düzeyinde artışların olduğu, ayrıca gebelik süresince artan LAP aktivitesinin doğumdan sonra tekrar azalmaya başladığı gözlendi. Sonuç olarak gebelik süresince ilave olarak verilen çinkonun LAP aktivitesinde herhangi bir değişikliğe neden olmadığı görüldü.
Abstract
Leucine aminopeptidase (LAP) has a zinc atom in its active site and this enzyme has important roles in the pregnancy. This study was aimed to investigate alterations on the activity of serum leucine aminopeptidase and the effect of zinc supplementation on the activity of leucine aminopeptidase in Tuj breed ewes during pregnancy. Twenty Tuj breed ewes at 3 years of age were used as animal material. The animals were divided into 2 equal groups including control (n=10) and the group receiving 30 mg/kg of zinc daily for average of 150 days (n=10). The blood samples from the groups were monthly collected and were centrifuged at 3000 rpm for 15 minutes. The activity of LAP in the serum samples were measured during the pregnancy (for five months) and after lambing. In this study, LAP activity between control and zinc-received groups was not significant. The LAP activity of both groups were found to be increased in the last third month of pregnancy. The activity of LAP increased during the last third month of pregnancy in both groups. In addition, the observed increase in LAP activity during the last 3 months of pregnancy declined again following the lambing. The results of this study showed that oral administration of zinc did not change the activity leucine aminopeptidase during the pregnancy compared to control.
