Effects of clomiphene citrate on the frequencies of sister chromatid exchange and mitotic index

Bu çalışmada infertilite problemi nedeni ile Celal Bayar Üniversitesi Tıp Fakültesi Kadın Hastalıkları ve Doğum Anabilim Dalı Polikliniği'ne başvuran ve ovulasyon indüksiyonu için Klomifen Sitrat uygulanan toplam 20 olguda, bu ilacın hücre düzeyinde oluşturabileceği akut mutajenik etkiler SCE ve MI açısından 3 ardışık siklus için değerlendirildi. Klomifen Sitrat tedavisinin hem mitotik indeks hem de kardeş kromatid değişimi üzerinde istatistiksel olarak anlamlı etkisi olduğu ve bu etkinin ilaç kesiminden üç ay sonra ortadan kalktığı gözlendi. Sonuç olarak, bir ovulasyon indüksiyon ajanı olan Klomifen Sitrat'ın olası genotoksik etkisi gözönüne alınarak yarar-zarar hesabının iyi değerlendirilerek uygulanması gerektiği düşünüldü.
Abstract
OBJECTIVE: To determine acute mutagenic effects of clomiphene citrate on sister chromatid exchange and Mitotic index. STUDY DESIGN: The study included 20 women using clomiphene citrate for three consecutive menstrual cycles. They were given 100 mg/day clomiphene citrate for 5 days between 3 and 7 days of menstrual cycles. Sister chromatid exchange and Mitotic index frequencies were recorded from peripheral blood leukocytes in a blinded fashion. RESULTS: There was statistically significant increase in SCE and Ml frequencies at the end of the third cycles (p<0.001). These increased rates reclined to normal level at the three months after stopping clomiphene citrate (p<0.001). CONCLUSION: Clomiphene citrate, which is an ovulation induction agent, might have a potential reversible mutagenic effect on cell division and DNA and this effect reclined to normal level after withdrawal of the drugs. The use of this drug is a risk of genotoxic damage and it is advisable to make a risk-benefit evolution in any particular case before its prescription .
