Vascular endothelial growth factor expression and clinicopathologic factors in epithelial ovarian carcinoma

Özet

Bu çalışmada vasküler endotelyal büyüme faktörü ekspresyonunun over karsinomundaki prognostik değeri araştırılmıştır.1990-1999 yılları arasında primer epitelyal over karsinomu tanısı almış 47 hasta (evre I-IV) çalışmaya dahil edilmiştir. "Borderline" over tümörleri ve primer peritoneal karsinomalar çalışmaya dahil edilmemiştir. Bütün hastalarda FIGO kriterlerine göre cerrahi evreleme ve evreye göre tümör "debulking" gerçekleştirilmiştir. İlk tümör örneklerinden 5 ìm kalınlığında slaytlar hazırlanarak vasküler endotelyal büyüme faktörü ile immünohistokimyasal olarak boyanmıştır. Daha sonra sonuçlar hasta yaşı, tümör evresi, histolojik tipi, tümör "grade"i, asit miktarı, tümör boyutu, preoperatif CA 125 düzeyleri, lenf nodu tutulum durumu, tümörün overleri tek taraflı veya bilateral olarak tutmuş olma durumu, postoperatif rezidü tümör ve hasta sağkalım süresi gibi klinikopatolojik faktörler ile karşılaştırılmıştır. VEGF ekspresyonu ile tanımlanan klinikopatolojik faktörler arasında arasında herhangi bir ilişki saptanmamıştır. Ancak kuvvetli VEGF ekspresyonu gösteren grupta asit miktarının daha yüksek olduğu saptanmıştır. VEGF daha önce de tanımlandığı gibi potent bir vasküler permeabilite faktörüdür ve epitelyal over kanserinde görülen asit ile ilişkili olabilir.

Abstract

OBJECTIVE: This study was set out to clarify the association between vascular endothelial growth factor (VEGF) expression status and clinicopathologic factors in primary epithelial ovarian carcinoma. STUDY DESIGN: Tumor samples of 47 patients with primary epithelial ovarian carcinomas (stages I-IV) those undergone surgical staging according to FIGO between 1990-1998 were evaluated for VEGF expression. 5 mm thick sections were prepared from the paraffin embedded tissues of the initial tumor specimens. Slides were stained immunohistochemically with VEGF by using "streptavidin biotin peroxidase" procedure. Results were correlated with clinicopathologic factors as age, tumor type, tumor grade, FIGO stage, lymph node involvement, ascite volume, tumor volume, patient survival, postoperative residue tumor, preoperative CA 125 levels, tumor unilaterality/bilaterality, and patient survival. RESULTS: There was not any correlation found between VEGF expression clinicopathologic factors. But importantly, we found a positive correlation between ascite volume and VEGF expression. In an other word we found ascite volume to be increased in those with strong VEGF expressions. CONCLUSION: As commonly known and well described, VEGF is a potent vascular permeability factor and may be related with increased ascite volume in epithelial ovarian carcinoma.
