HBsAg pozitif serumlarda HBV-DNA'nın iki ayrı PCR yöntemi ile taranması ve sonuçlarının serolojik göstergelerle karşılaştırılması

Özet
Bu çalışmada İnönü Üniversitesi Tıp Fakültesi Mikrobiyoloji Anabilim Dalı'nda HBsAg pozitif serum ör¬neklerinden rastgele seçilen 80 serumda iki ayrı Polimeraz Zincir Reaksiyonu (PCR) yöntemi ile HBV-DNA bakıldı. Pröteinaz-K /fenol ekstraksiyon yöntemi (I. yöntem) ile serumların 25'inde (%3I.3) HBV-DNA pozi¬tifliği tespit edildi! Ticari kit (Micro DNA izolasyon kiti, Strategene ve Digene Sharp Signal System kiti) kulla¬nılan II. yöntemde, serumların 23'ünde (%28.8) HBV-DNA pozitif bulundu, HBeAg pozitif olan 18 serumun hepsinde (% 100) her iki yöntemle HBV-DNA tespit edildi. Anti-HBe pozitif olan 62 serumun 7'sinde (% 11.3) I. yöntem ile HBV-DNA pozitif iken II. yöntem ile 5'inde (% 8.1) pozitif bulundu. İki yöntem sonuçlan arasın¬da anlamlı fark tespit edilemedi (p> 0.05). Sonuç olarak PCR laboratuvarları HBV-DNA tespitinde bu iki yöntemden kendi koşullarına uygun olan birini tercih edebilirler.
Abstract
Detection of HBV-DNA by Two PCR Methods in HBsAg Positive Sera, and Comparison of the Results with Serological Markers In this study, HBV-DNA was analysed by two different Polymerase Chain. Reaction (PCR) methods in randomly selected 80 HBsAg positive sera, which were previously detected as serologicaly HBV positive in Department of Microbiology of İnönü University School of Medicine. HBV-DNA positivity was found by proteinase-K/phenol method (method I) in 25 of 80 sera (31.3%). HBV-DNA was detected in 23 sera (28.8%) by method II in which commercial kit was used (microDNA isolation kit, Strategene and Digene Sharp Signal System kit). In all of 18 HBeAg positive sera (100%), HBV-DNA was detected by both of the methods. HBV-DNA positivity was detected in 7 of 62 anti-HBe positive sera (11.3%) and in -5 out of same samples (8.1%) by method I and II, respectively. There was no significant difference between the results of the two methods (p> 0.05).As a result, PCR laboratories can choose the methods appropriate for there facilities in detecting HBV-DNA.
