PET/BT Uygulamasının Sıçan Beyin Dokusunda TRPM2 ve Apoptozis Üzerine Etkileri

Özet

Amaç: Bu çalışma ile PET/BT uygulanan sıçanların beyin dokularında oksidatif hasarla etkileşimi bilinen Transient Reseptör Potansiyel Melastatin 2 (TRPM2) iyon kanallarının immünreaktivitesinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Çalışmada 20 adet Sprague Dawley cinsi dişi sıçan kullanıldı. Denekler 4 gruba ayrıldı. Grup I' deki sıçanlar kontrol olarak kullanıldı. Grup II' yi oluşturan sıçanlara deneyin ilk günü BT çekildi, grup III' teki sıçanlara deneyin ilk günü tek doz 1 mci2-floro-2-Deoksi-DGlukoz (FDG) uygulamasını takiben 1 saat sonra PET/BT çekildi. Grup IV' ü oluşturan sıçanlara ise yine deneyin birinci günü tek doz 1mci dozunda FDG uygulandı. Deney sonunda sıçanlar anestezi altında dekapite edildi ve beyin dokuları çıkartıldı. Rutin takipler yapılarak dokular parafin bloklara gömüldü. Bloklardan alınan kesitlere apoptozisin değerlendirilmesi için TUNEL boyama, TRPM2 ve Malondialdehit (MDA) ekspresyonları için avidin-biotin-peroksidaz yöntemi uygulandı. TUNEL ve immunohistokimyasal boyama sonrası dokular ışık mikroskobunda incelenerek fotoğraflandı ve istatistiksel analizleri yapıldı. Bulgular: Kontrol grubu ile karşılaştırıldığında diğer tüm gruplarda sıçan beyin dokularında apoptotik indeks ile TRPM2 ve MDA immunreaktivitelerinde istatistiksel olarak anlamlı bir farklılık izlenmedi. Sonuç: Bu çalışma ile PET/BT uygulanmasının sıçan beyin dokularında apoptozise neden olmadığı, TRPM2 iyon kanalları ve MDA ekspresyonları üzerine önemli bir etkisinin bulunmadığı kanaatine varıldı

Abstract

The Effects of PET/CT Application on TRPM2 and Apoptosis in Rat Brain TissueObjective: In this study it was aimed to examine the immunoreactivity of Transient Receptor Potential Melastatin 2 (TRPM2) ion channels which has a known interaction with oxidative damage in the brain tissues of rats undergoing PET/CT. Material and Method: In this study 20 adult Sprague Dawley female rats were used. The rats were divided in to 4 groups. Group I was used as control. On 1st day of experimental study computed tomography administered to the second group, to the third group a single dose 1mci FDG was injected and PET/CT was performed after 1 hour. To the fourth group the single dose of 1mci FDG was injected on 1st day of the experimental study. Rats were decapitated and brain tissues were removed. Routine follow up was performed and tissues were embedded in paraffin blocks. Sections taken from the blocks were administrated tunnel staining for apoptozis and avidin-biotin-peroxidase method for TRPM2 and MDA. Tissues were examined in the light microscope and photographed and then statistical analysis was performed. Results: When compared with the control group, in all other groups no significant differences were determined in brain tissues for apoptotic index, TRPM2 and MDA immunreactivities. Conclusion: In this study, we concluded that, PET/CT imagining doesn't cause apoptosis in rat brain tissue, moreover there is no significant effect on TRPM2 ion channels and MDA expressions
