Özet
Bu çalışmada, hepatit B virüsü pozitif hastaların serumundan HBsAggen bölgesi amplifiye edilmiş, bu amplikonlar önce Escherichia coli (E. colı) klonlama vektörüne aktarılmış daha sonra bu vektörden alınan gen bölgesi baculovirüsler için transfeksiyon vektörü olan pMelBac plazmidine klonlanmıştır. Elde edilen rekombinant vektör, böcek (Trichiplusia ni) hücre kültürüne (High Five Cell) transfekte edilmiştir. Daha sonra hücre kültürü içinden, rekombine olan hücreler seçilmiş ve HBsAg ürettikleri ELFA ile gösterilmiştir.
Abstract
In this study HBsAg genomic region of HBV was amplified from HBV positive patients sera. The amplified products, amplicons, were at first cloned to E coli cloning vector and then the genomic region which was taken from E. coli cloning vector was cloned to pMelBac plasmid to prepare transfection vector of baculovirus. The obtained recombinant vector was transfected to Trichiplusia ni cell culture. And then recombinant cells were selected from the cell culture and the production of HBsAg by these cells was displayed by ELFA.
