Analysis of the BACE1 and Clusterin Genes Expression Levels in Alzheimer’s Disease

Özet
Objective: This study aimed to explore the mRNA expressions of the β-site amyloid precursor protein-cleaving enzyme 1 (BACE1) and clusterin (CLU) genes in blood samples of patients with Alzheimer’s disease (AD) and healthy subjects. Methods: The expression levels of CLU and BACE1 in blood samples of subjects were examined using the real-time polymerase chain reaction (RT-PCR). The expression levels of patient and control groups were calculated using the 2-ΔΔCt method. The Statistical Package for Social Sciences (SPSS) version 22 (IBM Corp., Armonk, NY, USA) was used for statistical analysis, and a p of ˂0.05 was accepted as statistically significant. Results: The BACE1 and CLU genes were found 14-fold and 13-fold more expressed in the blood of the AD group than on the control group, respectively. Conclusion: Our study showed that the blood mRNA levels of the BACE1 and CLU genes were associated with AD.
Abstract
Amaç: Calışmamızda Alzheimer hastaları ve sağlıklı bireylerin kan örneklerinde β-amiloid öncü protein parçalayıcı enzim 1 (BACE1) ve clusterin (CLU) genlerinin mRNA ekspresyonunu araştırmayı amaçladık. Yöntemler: Kan örneklerinde Clusterin ve BACE1 ekspresyon seviyeleri Real Time polimeraz zincir reaksiyonu (RT-PCR) ile incelenmiştir. Hasta ve kontrol gruplarının ekspresyon düzeyleri 2-ΔΔCt yöntemine göre hesaplandı. İstatistiksel analiz için SPSS versiyon 22 (IBM Corp., Armonk, NY, ABD) kullanıldı ve p<0,05 istatistiksel olarak anlamlı kabul edildi. Bulgular: Alzheimer hasta grubunun kanlarında kontrol grubuna kıyasla BACE1 ve Clusterin gen ekspresyonları sırasıyla 14 ve 13 kat daha yüksek bulundu. Sonuç: Çalışmamızda BACE1 ve Clusterin genleri kan mRNA düzeylerinin Alzheimer hastalığı ile ilişkili olduğunu belirledik.
