Özet

Tecrübevi çalışma maksadı ile Ankara- Etlik BakteriyolojiEnstitüsü Şap Seroloji ve Doku Kültürü Laboratuvarında mavi dilaşısı hazırlanmıştır.Aşının hazırlanması için 14 çeşitli suç Meksikadan getirtilmiştir.Bu çalışmada 60 pasajla yumurta embriyosuna adapte edilmişMossap suşu kullanılmıştır.8 günlük embriyonl uyumurta sarısına (BTV) Mossap suşunun1/10 dilusyonundan 0,2 cc. inakule edilmiştir. İnokulasyondan sonra48 saat zarfında ölenler nazarı itibare alınmamıştır. Müşahade inakulasyondan sonra 5 gün devam etmiştir. İnokulasyondan sonra3 - 5 gün içinde ölen yumurtaların muhtevisi toplanmıştır. Bu yumurtamuhtevası aynı miktar Buffer- Laktoz- Pepton vasatı ile karıştırılmıştır. Herbir cc. için 100 ünite penicilline ve 100 mikrogramstreptomycin ilave edilmiştir.Bu karışım 0,75 cc. miktarında ampullere taksim edilmiş ve freeze-dryer'de kurutulmuştur. Aşı titrasyonu Reed ve Muench metodunagöre kurutulmadan evvel ve sonra olmak üzere iki defa yapılmıştır.Kurutmadan evvelki titrasyona 10-6,2 kurutmadan sonrakititrasyon 10-5,6 olarak tesbit edilmiştir.Aşının muafiyet denemeleri henüz yapılmamıştır.

Abstract

Blue Tongue vaccine was prepared at Foot and Mouth DiseaseSerology and Tissue Culture Laboratory of Etlik BaderiologyInstitute· Ankara, for experimental study. To prepare the vaccine 14 of several strains of the virus werebrought from Mexico. Mossap strain which had been suitably attenuated by 60 serial passages in embryonating chicken eggs wasused in this study.Fertile eggs, preincubated for 8 days, were inoculated with0,2 ml of 1/ 10 dilution of Mossap strain via yolk sac route. Death ofembryos duringthe first 48 hours after inoculation, were discarded.Observation continued for 5 days. The contents of the eggs ofembryos which haddiedwithin 3th. to 5th. days postinoculation,were collected. The contents of the eggs were mixed with equalvolume of Buffer- Lactose- Pepton medium, 100 units of penicillineand 100 microgram streptomycin per milliter were also added.The mixture was dispensed in 0,75 ml quantities into each ampuleand desiccated in Freeze- Dryer- appartus. The titration ofthe vaccine examined by the method of Reed and Muench beforeand after desication. It was calcullated that the titration beforedesiccation was 10-6,2 after desiccation was 10-5,6
