Abstract

The Protective Effect of Radical Scavenger and Anti-oxidant on Gastric Mucosa Injured by Ethanol The aim of this study was to investigate the effect of absolute ethanol at high doses on gastric injury, and to exa¬mine lipid peroxide levels and histological effects on gastric tissues in rats. The protective effect of such radical scavenger agents as a allopurinol, sucralphate and anti-oxidant vitamin E on gastric damage were also investigated. Forty-five wistar albino rats, each weighing 180-200 gr were used. Experimental animals were equally divided into 5 groups, as following; Group-L control group, Group-IL Ethanol, Group-Ill: Vit E + ethanol, Gro-up-VI: Allopurinol+ethanol, Group-V: Sucralphate+ethanol. The control group was sacrifced without any expe¬rimentation, while 1 ml of '96% ethanol was administered to ethanol group by orogastric following 24 hours of starvation. Allopurinol, sucralphate and vitamin E were administered to other group for one week. After the ex¬perimental period, the animals were sacrificed under ether anesthesia, and their gastric tissues were removed by systemic perfusions. The tissues were then fixed with 10% neutral formalin solution (pH: 7.4). Five pim pa-rafine sections were taken routinely and stained with Hematoxylen-Eosin and later observed under light mic-roscobe. 0.5 gr of tissue dissected from this materials was used to determine Malondialdehyde (MDA) levels in all groups. MDA levels, an indicator of tissue lipid peroxidation were found to be 85-4+17.7 nmol/g in control group, 190.7+16.7 nmol/g in ethanol group, 91-9+-15- 7 nmol/g in Allopurinol+ethanol group, 79.6+15.4 nmol/g in Vit E+ethanol group and 131-9±16.4 nmol/g in sucralphate+ethanol group. As a result, gastric mucosal ero¬sions and lipid peroxide levels were observed to increase in ethanol and sucralphate+ethanol groups, compared to control group. In particular, radical scavenger agents such as vit E, allopurinol and sucralphate were found to reduce lesion formation at histological and microscopical level, and to be protective for lipid peroxidation for¬mation. Sucralphate, however, was found to be less effective respect to these agents.

£ah§mada, yuksek dozlarda mide hasan olu§turan absolu etanolun, siganlann mide dokulannda lipiddperok- sit duzeyleri ve olu§an hasarin histolojik incelemesi ile radikal temizleyici ajan allopurinol, sukralfat ve anti- oksidan E vitamininin bu hasara kar§i etkinliginin ara§tinlmasi amaglanmi§tir. Bu amagla 45 adet 180-200 gr agirhginda Wistar albino sigan kullanildi. Deney hayvanlan e§it olarak 5 gruba boliindu. Grup I: Disney hayvanlan kontrol grubu olarak, Grup IL Etanol, Grup III: Vit E + etanol, Grup VI: Allopurinol + etanol, Grup V: Sukralfat + etanol gruplan olarak kullanildi. Kontrol grubuna higbir giri§im yapilmadan sakrifiye edildi. Etanol grubuna 24 saat aghgi takiben 1 ml % 96'hk etanol orogastrik sonda ile uygulandi. Diger her bir gru¬ ba bir hafta sureyle allopurinol, sukralfat ve vitamin E verildi. Deney hayvanlannin deney sonunda, eter anes- tezisi altmda sistemikperfuzyonlanyapilarak mide dokulan ahndi ve %10'luk notralformalin (pH:7.4) soliis- yonunda fiksasyonu yapildi. Rutin histolojik takiplerle elde edilen 5 mm kahnhgmda parafin kesitleri Hemo- toksilen-Eozin ile boyanarak isik mikroskobik degerlendirilmesi yapildi. Kalan materyalden her bir grup igin 0.5 gr mide dokusu ahndi vebutun deney gruplannda malondialdehit (MDA) tayini yapildi. Qah§ma sonu- cunda doku lipid peroksidasyonunun bir gostergesi olan MDA miktan, kontrol grubunda 85.4 ± 17.7 nmol/gidi. Sadece etanol verilen grupta 190.7+ 16.7 nmol/g iken, allopurinol + etanol grubunda 91.9 ± 15.7 nmol, vit E + etanol grubunda 79-6 + 15.4 nmol/g, sukralfat + etanol grubunda ise 131.9 ± 16.4 nmol/g idi. Sonug olarak, etanol ve sukralfat + etanol uygulanan grupta kontrol grubuyla karsilastinldigmdamide mukoza erozyonlannin ve lipidperoksit dilzeylerinin arttigi gozlendi. Ozellikle vit E, allopurinol ve sukralfat ileprofilaksi yapilan gruplar sadece etanol verilen grup ile karsilastmldiginda radikal giderici ajanlann hem makroskobik hem histolojik dilzeyde hem de lipidperoksidasyonu yonunden koruyucu olduklan gozlendi. Bunlara gore sukralfat daha az etkiliydi.
