Effects of selenium and vitamin E on glucose, lipid values, platelet indices and renal tissue in experimental diabetes

Abstract

Objective: Diabetic agents increasing free radicals may cause oxidative damage in tissues and affects platelet indices. Vitamin E and selenium (Se), which are important antioxidants, may protect harmful effects of the free radicals. In this study, it was aimed to investigate the effects of Se and vitamin E on some biochemical parameters, platelet indices and renal tissue in experimentally induced diabetes. Material and Methods: This study was performed on Wistar albino rats. The first group was used as a control. The second group was used as diabetes injected 65 mg/kg of streptozotosin (STZ). The third group received with Se (Na2SeO3, 0.3 mg/kg body weight), the fourth group with vitamin E (dl-α-tocopheryl acetate, 100 mg/kg body weight), and the fifth group with Se plus vitamin E combination (Na2SeO3, 0.3 mg/kg body weight+dl-α-tocopheryl acetate, 100 mg/kg body weight) for every other day intervals during fifteen days. Blood and kidney tissue samples of all rats were analyzed 15 days after STZ injection. Results: The values of glucose, trigliserid, cholesterol and low density lipoprotein were increased in diabetes group; plateletcrit and platelet counts were decreased. The correlation between mean platelet volume and cholesterol was significant in only diabetes group. The levels of glucose, low density lipoprotein and amylase activity were decreased by treatment of the antioxidants. Renal tissues were not affected in diabetes and antioxidant groups. Conclusion: Se and vitamin E may affect on some platelet indices, glucose concentration and lipid values. Se and vitamin E supplementation in diabetic rats may induce hypolipidemic and hypoglycemic effects, and these antioxidants may be used for prophylactic purpose in diabetes.

Amaç: Diyabetik ajanlar serbest radikalleri artırarak dokularda oksidatif hasar oluşturabilir ve trombosit indekslerini etkileyebilir. Eksojen antioksidanlardan E vitamini ve selenyum (Se), serbest radikallerin zararlı etkilerini azaltabilir. Bu çalışma, deneysel diyabette bazı biyokimyasal değerler, trombosit indeksleri ve böbrek dokusu üzerinde E vitamini ve Se’un etkilerini araştırmak amacı ile yapıldı. Gereç ve Yöntem: Çalışma Wistar albino sıçanlar üzerinde yürütüldü. Birinci gruptaki sıçanlar kontrol olarak kullanıldı. İkinci gruptaki sıçanlarda 0.1 M sitrat tamponunda hazırlanmış 65 mg/kg streptozotosin (STZ) intraperitoneal (ip) olarak uygulanarak diyabet oluşturuldu. STZ enjeksiyonuna ilaveten üçüncü gruba Se (sodyum selenat 0.3 mg/kg), dördüncü gruba E vitamini (alfa-tokoferol asetat, 100 mg/kg), beşinci gruba ise Se-E vitamini kombinasyonu (sodyum selenat 0.3 mg/kg+alfa-tokoferol asetat, 100 mg/kg) gün aşırı, on beş süreyle uygulandı. STZ enjeksiyonundan 15 gün sonra tüm sıçanların kan ve böbrek dokusu örnekleri alınarak incelendi. Bulgular: Diyabet grubunda ortalama trombosit hacmi ile kolesterol değerleri arasında önemli ilişkiler olduğu; glukoz, trigliserit, kolesterol ve düşük dansiteli lipoprotein, değerlerinin yükseldiği, ancak trombosit sayısı ve trombokrit değerlerinin azaldığı, Se veya E vitamini-Se’un kombine verilmesi ile glukoz ve düşük dansiteli lipoprotein değerleri ile amilaz aktivitesinin azaldığı gözlemlendi. Kontrol ve deneme gruplarında böbrek dokusunda ışık mikroskobu düzeyinde önemli bir değişiklik gözlenmedi. Sonuç: E vitamini ve Se’un bazı trombosit indeksleri ile glukoz değerleri ve lipitleri etkileyebileceği belirlendi. Bu sonuçlara göre E vitamini ve Se uygulaması, deneysel diyabette hipoglisemik ve hipolipidemik etkiler oluşturabilir; bu antioksidanlar diyabette profilaktik amaçla kullanılabilir.
