Cytokine response of mononuclear cells against staphylococcus epidermidis-extracted slime substance

Abstract
Certain strains of Staphylococcus epidermidis produce a mucoid slime that appears to be an important virulence factor. It was reported that slime has several inhibitory effects on immune effector functions such as phagocytosis of neutrophils, lymphoproliferative response and NK cytotoxicity. To test whether inhibitory effects of slime on immune system has any relationship to the induction of cytokine types from mononuclear cells, its effect on the cytokine induction from mononuclear cells was measured. Mononuclear cells were stimulated with varying concentrations (3, 30 and 300 mg/ml) of S. epidermidis-extracted slime substance. After a 72-h incubation period it was observed that slime substance did not induce IL-4 and IL-12 production from mononuclear cells. But it induced IFN-y, IL-6 and IL-10 production from mononuclear cells.
Özet
Staphylococcus epidermidis'in belirli suşlan önemli bir virulans faktörü olarak görünen bir mukoid slime üretir. Slime maddesinin nötrofil fagositozu, lenfoproliferatif cevap ve NK sitotoksisitesi gibi immün effektör fonksiyonlar üzerinde bazı önleyici etkilerinin olduğu bildirilmiştir. Slime maddesinin immün sistem üzerindeki inhibitor etkisiyle mononükleer hücrelerden çeşitli sitokinlerin salınımı arasındaki ilişkiyi araştırmak için, slime maddesinin mononükleer hücrelerden sitokin salgılanmasına etkisi araştırıldı. Mononükleer hücreler değişen konsantrasyonlardaki (3, 30 ve 300 ug/ml) S. epidermidis'den elde edilen slime maddesi ile uyarıldı. Yetmişiki saatlik inkübasyon süresi sonunda slime maddesinin mononükleer hücrelerden IL-4 ve IL-12 salgılanmasına neden olmadığı saptandı. Ancak mononükleer hücrelerden IFN-y, IL-6 ve IL-10 salgılanmasına neden olmaktadır.
