Toxic and behavioural effects of Tanacetum abrotanifolium L. DRUCE (Asteraceae) stem and flower extracts on Sitophilus granarius and Sitophilus oryzae (Col., Curculionidae)

Abstract
Toxic and behavioral effects of Tanacetum abrotanifolium (L.) DRUCE (Asteraceae) stem and flower extracts were tested on Sitophilus granarius (L.) and S. oryzae (L.) (Col, Curculionidae) under laboratory conditions. The stem and flower extracts were obtained using three different solvents that were hexane, ethyl acetate and methanol. Both stem and flower extracts of T. abrotanifoliıum produced very high level of contact toxicity to the granary weevil. The greatest contact toxicity to granary weevil was seen with the stem hexane extract which caused 64% mortality. Dose-response bioassay with granary weevil exhibited that stem extracts were more toxic than the flower extracts and LD50 values were 5.54, 4.81 and 5.01 μg/insect for stem hexane ,ethyl acetate and methanol extracts respectively. In the second series of experimets, deterrent and antifeedant effects of plant extracts were tested with choice and non-choice set up. All tested plant extracts caused reduction in feeding of both S. oryzae and S. granarius. T. abrotanifolium extracts showed high level of deterrent activity to both species and their activity increased as the incubation time was extended. In choice set up, the most pronounced antifeedant activity was seen with stem methanol extract (71%) for the granary weevil and with flower-hexane extract (69%) for the rice weevil. In non choice set up, flower-hexane and flower-ethyl acetate extracts yielded the highest antifeedant activity (47%) against S. granarius and flower-hexane extract (71%) was the most active extract for S. oryzae. The results indicate that T. abrotanifolium has a potential in control of S.granarius.
Özet
Bu çalışmada Tanacetum abrotanifolium (L.) DRUCE’un gövde ve çiçek ekstraktlarının iki önemli depo zararlısı Sitophilus granarius (L.) ve S. oryzae (L.) (Coleoptera, Curculionidae)’ye karşı kontakt toksisiteleri uzaklaştırıcı ve beslenmeyi engelleyici etkileri laboratuvar şartlarında değerlendirilmiştir. Bitkinin gövde ve çiçek ekstraktları, farklı çözücüler olan hekzan, etil asetat ve methanol kullanılarak elde edilmiştir. T. abrotanifolium’un hem gövde hem de çiçek ekstraktları buğday biti üzerinde yüksek derecede kontakt toksisite göstermiştir. Tek doz tarama testlerinde buğday biti üzerinde en yüksek kontakt toksisiteyi % 64’lük öldürme oranı ile gövde hekzan ekstraktı göstermiştir. S. oryzae, bitki ekstraktlarına daha az duyarlılık göstermiş olup, en yüksek kontakt toksisite %9 ölüm oranı ile etil asetat gövde ekstraktında saptanmıştır. Buğday biti ile yapılan doz-ölüm denemelerinde gövde ekstraktlarının daha ektili olduğu belirlenmiş ve LD50 değerleri hekzan, etil asetat ve metanol ekstraktları için sırasıyla 5.54, 4.81 ve 5.01 μg/böcek olarak hesaplanmıştır. Çalışmanın ikici kısmında, bitki ekstraktlarının uzaklaştırıcı etkisi seçenek testleri ile beslenmeyi engelleyici etkileri seçenek ve zorunluluk testleri ile denenmiştir Yapılan çalışma sonuçları ile bitki ekstraktlarının her iki türde de yüksek oranda uzaklaştırıcı etkisinin olduğu ve etkinliklerinin zaman içerisinde arttığı saptanmıştır. Çalışmada kullanılan tüm bitki ekstraktları S. granarius ve S. oryzae’ nın beslenmesinde azalmaya neden olmuşlardır. Seçenek testlerinde en yüksek beslenmeyi engelleyici etki Buğday biti için % 71 ile gövde-metanol ekstraktından, Pirinç biti için % 69 ile çiçek-hekzan ekstraktından saptanmıştır. Zorunluluk testlerinde ise en yüksek aktivite Buğday biti için % 47 ile çiçek-etil asetat ve çiçek-hekzan ekstraktından Pirinç biti için %71 ile çiçek-hekzan ekstraktından elde edilmiştir. Çalışma sonuçları T. abrotanifolium’ un S. granarius ile mücadelede kullanma potansiyelinin olduğunu ortaya koymuştur.
