The determination of ochratoxin A in raisin and fig by high performance liquid chromatography

Abstract

A rapid, economic and reliable method is applied to determine ochratoxin A in raisin and figs. Ochratoxin A is extracted by 0.1 M H3PO4 and chloroform from raisin and figs. The extract is cleaned up using a Sep-Pak silica cartridge. For the figs which has more interferences, extra steps are added to cleaning procedure. Ochratoxin A is detected by HPLC and confirmed by TLC. The detection limit is 0.03 ppb,the recovery is 93.5-100 % in HPLC.

Özet

Üzüm ve incirde okratoksin A'nın saptanması için hızlı, ekonomik, güvenilir bir metot uygulanmıştır. Okratoksin A, üzüm ve incirden 0.1 M H3PO4 ve kloroform ile ekstre edilmiş ve ekstrakt tek kullanımlık Sep-Pak silika kartuş ile temizlenmiştir. Girişim yapan maddelerin fazla olduğu incirde ise temizleme basamağına ilaveler yapılmıştır. Okratoksin A, HPLC ile saptanmış ve TLC'de doğrulanmıştır. HPLC ile ölçümlerde minimumsaptama limiti 0.03 ppb ve geri kazanım% 93.5-100'dür.
