Heterografting of ovarian cortex tissue pieces heterotopically to ductus deferens and cortex in testes of sheep

Abstract
Objective: We'aimed to search and compare the development and survival of ovarian follicles after heterotopically transplanted to ductus deferens which is protected and testicular cortex which is unprotected against immune system and thus we wanted to investigate the probability of conserving the ovarian follicles of women having the risk of loosing their ovarian function because of chemotherapy and/or radiotherapy required for malignancies at early ages. Institution: Yuzuncu Yil University Veterinary Medicine Faculty Research Laboratory. Material and Methods: In this study, one-year-old five male and one female sheep were included. Bilateral oophorectomy was performed through a laparo-tomic operation. Ovarian cortex was divided into 1x2 mm. multiple tissue sections. The ovarian tissue sections was placed into ductus lumen in the left testes of all male sheep. At the same time, the tissue samples taken from ovarian cortex was implanted within testicular cortex. Five months later, tissues taken from the left testes were histopathologically examined. Results: Normal ovarian tissue was not encountered at the end of the examination of ductus deferenses of five male sheep's testes. Fibrosis and calcification were seen. Examination of ovarian tissues implanted within testicular cortex revealed some areas of calcification and remnants of primary follicles in the site of implantation within the testicular cortex of one sheep. The implantation site in the testicular cortex of the other four sheep, no ovarian tissue was found. Areas of calcification, hyalinization., fibrosis and multiple histiocytes were seen. Conclusion: As well as rejection of ovarian tissues implanted to ductus deferens may be resulted from the surgical procedure that makes ductus deferens open to the immune system which is normally protected against, High androgenic activity within ductus deferens'may play a role in atresia and rejection, too. We think that with more sophisticated studies, heterotopic ovarian cortex transplantation into ductus deferens can be successfull provided that ductus deferens protected from immune system and androgenic activitiy in Leydig cells decreased by hormonal supression.

Amaç: İmmün sistemden korunan duktus deferens ve immün sistemden korunmayan testis korteksine over foliküllerinin heterotopik olarak transplantasyonu sonrası folikül yaşamını ve gelişimini inceleyerek karşılaştırmak ve genç yaşta malignensisi olup, kemoterapi ve radyoterapi ile over fonksiyonunu kaybetme riski taşıyan kadınların over foliküllerinin eşin duktus deferensinde saklanabilme olasılığını araştırmayı amaçladık. Çalışmanın Yapıldığı Yer: Yüzüncü Yıl Üniversitesi Veterinerlik Fakültesi Araştırma Laboratuarı. Materyal ve Metod: Çalışmada; bir yaşında beş erkek ve bir dişi koyun kullanıldı. Dişi koyuna laparatomi ile bilateral ooferektomi yapıldı. Over korteksi 1x2 mm boyutunda çok sayıda doku kesitlerine bölündü. Tüm erkek koyunların sol duktus deferens lümenine over doku kesiti yerleştirildi. Aynı zamanda over korteksinden alınan doku örneği testis korteksinin içerisine implante edildi. 5 ay sonra sol testisten alınan dokular histopatolojik olarak değerlendirildi. Bulgular: Beş erkek koyun testisinin duktus deferenslerinin incelenmesinde; normal over dokusu izlenmedi. Fibrozis ve kalsifikasyonlar görüldü. Testis korteksine implante edilen over dokularının incelemesinde; bir koyunun testis korteksi implantasyon bölgesinde kalsifikasyon alanları ve primer folikül kalıntıları görüldü. Diğer dört koyunun testis korteks implantasyon bölgesinde over dokusu izlenmedi, kalsifikasyon alanları, hiyalinizasyon, fibrozis ve multipl histiositler izlendi. Sonuçlar:Duktus deferense implante edilen over dokularının rejeksiyona uğraması, immün sisteme kapalı olan duktus deferensin cerrahi işlemle immüniteye açık hale gelmesinden kaynaklanabileceği gibi, duktus deferensteki yüksek androjenik aktivite de atrezi de rol oynamış olabilir. Daha sofistike çalışmalar ile; duktus deferensi immunite etmeden ve leydig hücrelerinde hormonal süpresyonla androjenik aktiviteyi azaltarak, duktus deferenslere yapılacak over korteksinin heterotopik transplantasyonunun başarılı olabileceğini düşünmekteyiz.
