Abstract

This study was developed to investigate the effect of the ratio of citrate to blood on prothrombin time analysis. In addition, the results of the studies of the samples known with various citrate contents ran at once and after 4-hours separately were examined with respect to time dependency. 45 healthy male individuals (23±4 years old) were included in the study and venous blood samples were collected according to four groups which had ratios of 0. 1/1.9 -0.2/1.8 - 0.3/1.7 - 0.4/1.6 of citrate / blood, respectively. Prothrombin time assays of all samples were run immediately and after 4-hours in duplicate. All of the groups when compared to each other, had statistically significant differences for the immediate measurements. On the other hand, prothrombin time results of samples ran after 4-hours were found similar to those obtained immediately. As a result, it is suggested that it is necessary to pay attention for sampling in order not to obtain falsely increased prothrombin time values. It is also concluded that preanalytical errors should be detected when results, which are not correlated with the clinical signs and symptoms, are obtained. It also became evident that prothrombin time results remained unchanged even the samples were left for four hours without analyzed.

Özet

Bu çalışma, protrombin zamanı (PTZ) testinin, sitrat/kan (1/9) oranındaki değişikliklerden etkilenip etkilenmediğini incelemek üzere planlandı. Ayrıca, farklı sitrat oranlarına sahip örnekler hemen ve dört saat sonra okunarak, sonuçların zamana bağlı olarak değişip değişmediği araştırıldı. Çalışmaya 45 sağlıklı erkek birey katıldı, her bireyden alınan venöz kan örnekleri sitrat/kan olarak 1.grup 0.1/1.9 oranında, 2.grup 0.2/1.8 oranında, 3.grup 0.3/1.7 ve 4.grup 0.4/1.6 oranında hazırlandı. Tüm örneklerden, alınır alınmaz ve dört saat sonra PTZ testi ikişerli olarak çalışıldı ve dört grubun sonuçları birbirleriyle karşılaştırıldı. Hemen yapılan ölçümlerde tüm grupların sonuçları arasında istatistiksel anlamlı bir farklılık bulundu (p0.05). Sonuç olarak, yanlış yüksek PTZ değerlerinden kaçınmak için örnek alımına mutlaka dikkat edilmesinin gerekli olduğu ve klinikle uyumsuz, sonuçlarla karşılaşıldığında öncelikle preanalitik hataların gözden geçirilmesinin uygun olacağı düşünüldü. Ayrıca dört saate kadar beklemiş örneklerde, PTZ sonuçlarının değişmediği değerlendirildi.
