Investigation of the protective role of omega-3 fatty acids riched fish oil on rats given diethylnitrosamine

Abstract

This study aimed to determine toxic effects of diethylnitrosamine on liver and to investigate the protective effect of ω-3 fatty acids to oxidative stress related with the toxic effects of diethylnitrosamine. Fourty male rats weighing 212±6,41 g (approximately 4 months-old) were used in this study. The animals were equally divided into four groups, control (0,9 % NaCl solution intraperitoneally with a single dose), group II (150 mg/kg diethylnitrosamine intraperitoneally with a single dose), group III (150 mg/kg diethylnitrosamine intraperitoneally with a single dose and 0,4 mg/kg/day subcutaneus dose of fish oil containing ω-3 fatty acids) and group IV (0,4 mg/kg/day subcutaneus dose of fish oil containing ω-3 fatty acids). After seven-day-application, the blood was collected from heart under anesthesia and the liver samples were collected after euthanasia and preserved in 10 % formaldehyde. Part of blood samples were stored without any process as a whole blood. The plasma was seperated from remaining blood samples. The reduced glutathione in whole blood and level of malondialdehyde and activity of adenosine deaminase in plasma were colorimetrically determined. While the activity of adenosine deaminase and whole blood glutathione level in groups II and III increased comparing to that of in groups II, it was observed that plasma malondialdehyde level increased in group II comparing to that of control group. The results of this study may suggest the possibility of increase of oxidative stress related with toxic effects of diethylnitrosamine and protective effect of ω-3 fatty acids to this stress by blocking lipid peroxidation, increasing antioxidant molecules such as reduced glutathione and playing a role like scavenging of free radicals. Further study is needed on this subject since the activity of adenosine deaminase caused by diethylnitrosamine was not effectively prevented by ω-3 fatty acids in the current study.

Özet

Çalışmada, dietilnitrozaminin karaciğer üzerindeki toksik etkilerinin belirlenmesi ve bu toksik etkilere bağlı olarak meydana gelebilecek oksidatif strese karşı omega-3 (ω-3) yağ asitlerinin koruyucu etkisinin araştırılması amaçlanmıştır. Materyal olarak yaklaşık 4 aylık ve 212±6,41 g olan 40 adet Wistar albino cinsi erkek rat kullanılmıştır. Ratlar, kontrol (periton içi tek doz % 0,9’luk NaCl çözeltisi), grup II (periton içi tek doz 150 mg/kg dietilnitrozamin), grup III (periton içi tek doz 150 mg/kg dietilnitrozamin ile derialtı 0,4 g/kg/gün ω-3 yağ asiti içeren balık yağı) ve grup IV (0,4 g/kg/gün derialtı ω-3 yağ asiti içeren balık yağı) olmak üzere dört eşit gruba ayrılmıştır. Yedi gün süren uygulamanın sonunda anestezi altında kalplerinden heparinli tüplere kan örnekleri alındıktan sonra öldürülen ratların karaciğer dokuları çıkarılarak %10’luk formaldehitte saklanmıştır. Alınan kan örneklerinden bir miktarı hiçbir işleme tabi tutulmadan tam kan olarak ayrılmıştır. Geriye kalan kanların plazmaları elde edilmiştir. Tam kanda redükte glutatyon, plazmada malondialdehit düzeyi ile adenozin deaminaz aktivitesi kolorimetrik olarak tayin edilmiştir.
