Abstract

Objective: G?h (tissue transglutaminase; TGII), known as the ?1B-adrenoceptor signal mediator, is a bifunctional enzyme possessing transglutaminase and guanosine triphosphatase activities. The microtubule-associated protein tau plays a role in formation of neurofibrillary tangles in Alzheimer's disease (AD) brain. Tau is also known as a substrate of transglutaminase. We aimed to better understand the correlation between ?1B-adrenoceptor signaling and AD.Methods: We examined not only the cross-linking ability of TGII for tau, but also the expression level of tau as well as ?1B-adrenoceptor signaling molecules, TGII and phospholipase C-?1, in the human brain.Results: When the tau protein was assayed as a transglutaminase substrate of TGII, tau proteins formed cross-linked products. However, phospholipase C-?1 inhibited transglutaminase activity in TGII to cross-link with tau in vitro. The amount of expressed mRNA in AD brain tissue was elevated 2~10 fold for tau and 3~20 fold for TGII. Consistent with these observations, the densities of expressed proteins in AD brain tissue also increased 9 fold for tau and 15 fold for TGII. Moreover, phospholipase C-?1, which is a negative regulator for transglutaminase activity of TGII, also increased 2~25 fold for mRNA as well as 8 fold for protein in AD brain tissue. In contrast, expressed mRNA and protein activity for ?1B-adrenoceptor were almost the same between AD and normal brains. Conclusion: These results suggest that the ?1B-adrenoceptor signal mediator, TGII and tau, as a substrate of TGII might have a pathophysiological role in AD, but this correlation may be independent of the ?1B-adrenoceptor signaling system, including ?1B-adrenoceptor and phospholipase C-?1.

Amaç: ?1B-adenoseptör sinyal mediyatör olarak bilinen G?h (doku transglutaminaz; TGII),transglutaminaz ve guanizin trifosfataz aktiviteleri içeren çift fonksiyonlu bir enzimdir. Alzheimer hastalığı (AD) olan beyin dokusunda nörofibriller yumakların oluşması için mikrotübülüs ilişkili tau proteini rol oynamaktadır. Bu protein (tau) aynı zamanda transglutaminazın substratıdır. Bu makalede ?1B-adrenocseptör sinyali ve AD arasındaki ilişkinin daha iyi anlaşılması amaçlanmıştır.Metod: Bu çalışmada, tau proteini için yalnızca TGII'nin çapraz-bağlanma becerisini değil, insan beyninde tau ile ?1B-adrenoceptor sinyal molekülleri, TGII ve phospholipase C-?1 ekspresyon seviyeleri de incelenmiştir.Bulgular: Tau proteini TGII'nin transglutamiaz substratı olarak test edildiğinde çapraz bağlı ürünler oluşturmuşlardır. Ancak, in vitro çalışmada, fosfolipaz C-?1, TGII'deki transglutaminazın tau ile çapraz bağlanmasını inhibe etmiştir. AD olan beyin dokusunda eksprese edilen mRNA miktarı, tau için 2-10 katı, TGII için ise 3-20 kat artış göstermiştir. Bu bulgular ile uyumlu olarak AD olan beyin dokuda eksprese olan prtein yoğunlukları tau için 9 katı, TGII için ise 15 katı artmıştır. Bunlara ek olarak, TGII'nin transglutaminaz aktivitesinin negatif regülatörü olan fosfolipaz C-?1'de mRNA için 2-25 kat AD hastalığı olan beyin dokusu içinse 8 kat artış göstermiştir. Buna karşılık eksprese mRNA ve ?1B-adrenoseptör protein aktiviteleri normal ve AD olan beyin dokularında aynı kalmıştır.Sonuç: Bu sonuçlara göre ?1B-adrenoseptör sinyal mediyatörü olan TGII ve TGII ve substratı olan tau AD'de patofizyolojik role sahipler ancak bu korelasyon, ?1B-adrenoseptör ve fosfolipaz C-?1 de dahil olmak üzere ?1B-adrenoseptör sinyal sisteminden bağımsızdır.
