Increased serum levels of spectrin degradation products in patients with schizophrenia

Abstract

Objective: Under various patho-physiological and physiologicalconditions, spectrin breakdown reactions generateseveral spectrin breakdown products of 120 kDa(SBDP120) and 145 kDa (SBDP145). Previous studies indicatingthat there is the existence of a raised breakdownof α-spectrin in schizophrenic left superior temporal cortices.In this study, we aimed to investigate serum levelsof SBDP120 and SBDP145, which has not been previouslyexamined, and investigate their relationships with clinicalparameters in patients with schizophrenia.Methods: Forty-four patients with schizophrenia, followedby psychotic disorders unit, and 44 healthy controls, ageand gender-matched volunteers with no psychiatric history,were included in this study. Sociodemographic form wasapplied to both groups. Turkish version of positive andnegative syndrome scale (PANSS) were implemented to thepatients. Serum SBDP120 and SBDP145 levels were determinedby Enzyme-Linked Immuno Sorbent Assay.Results: Serum SBDP120 ng/mL and SBDP145 ng/mLlevels of the patients with schizophrenia were significantlyhigher than healthy controls. Even more important,serum SBDP120 levels were positively correlated withPANSS scores in patients with schizophrenia.Conclusions: These findings may provide evidence fordisturbance of neuroplasticity, membrane/cytoskeletonstability, dynamics, and remodelling in schizophreniapatients and support the neurogenerative theories forexplaining the etiology of schizophrenia.

Özet

Amaç: Değişik pato-fizyolojik ve fizyolojik koşullarda, spektrin yıkılım reaksiyonları, çeşitli 120 kDa (SBDP120) ve 145 kDa (SBDP145) spektrin bozunum ürünlerini üretir. Önceki çalışmalar şizofrenide sol superior temporal kortekste α-spektrin düzeyinde artmış bir yıkılım olduğuna işaret etmektedir. Bu çalışmada, şizofreni hastalarında daha önce incelenmemiş olan SBDP120 ve SBDP145’in serum düzeylerini ve klinik parametrelerle ilişkisini araştırmayı amaçladık. Metod: Psikotik bozukluk birimi tarafından takibi yapılan 44 şizofreni hastası ve yaş ve cinsiyetce eş, psikiyatri öyküsü olmayan 44 gönüllü sağlıklı kontrol olarak çalışmaya dahil edildi. Her iki gruba sosyodemografik form uygulandı. Pozitif ve negatif sendrom ölçeğinin Türkçe versiyonu hastalara uygulandı. Serum SBDP120 ve SBDP145 seviyeleri Enzim-Bağlı İmmunosorbent Testi ile belirlendi. Bulgular: Şizofreni hastalarının serum SBDP120 ng/mL ve SBDP145 ng/mL düzeyleri, sağlıklı kontrollerden anlamlı derecede yüksekti. Daha da önemlisi, şizofreni hastalarında serum SBDP120 düzeyleri pozitif ve negatif sendrom ölçeği (PANSS) skorlarıyla pozitif yönde ilişkiliydi. Sonuç: Bu bulgular, şizofreni hastalarında nöroplastisite, membran/hücre iskeleti stabilitesi, dinamikleri ve yeniden şekillenmesindeki bir bozukluğa ilişkin kanıt sağlayabilir ve şizofreninin etiyolojisini açıklayan nörodejeneratif teorileri desteklemektedir.
