Synthesis of 2-aminonaphthalene- 1-sulfonic acid Schiff bases and their interactions with human serum albumin

Abstract
Objectives: Human serum albumin (HSA) can bind andtransport many substances to cells to meet various needs ofthe organism. The binding efficacy of HSA to these substancesdirectly affects their functions. In this paper twoSchiff base compounds were synthesized to explore theinteraction between HSA and both compounds.Methods: Fluorescence spectra and an AutoDock modelwere utilized to investigate the interaction mechanismand binding model between proteins and Schiff baseproducts. The conformation change of HSA was detectedby resonance light scattering and circular dichroismspectra.Results: The two compounds bound easily with HSA, withbinding constants of 104. The binding sites for both compoundsin HSA were within an appropriate distance forlong-range interactions. Both compounds are accommodatedin hydrophobic domains of HSA. However, electrostaticinteractions and other supermolecular forces coexistbetween the compounds and protein. Binding of thesecompounds disturbed the protein secondary structure andcaused a certain degree of destabilization.Conclusions: The two Schiff base compounds can interactwith HSA with high efficacy, which is helpful for explorethe application of this type of Schiff base in biomedicalresearch.
