Abstract

Aim: The aim of this study is to investigate the effect of systemic (intraperitoneally) two different doses ondansetron administration on analgesic activity of bupivacaine administered intrathecally in rats.Materials and Methods: 24 rats were included in the study. Group Saline (Group S) (n=6): 10 µL of saline was administered intrathecally (IT) and 0.5 mL of saline was administered intraperitoneally. Group Bupivacaine (Group B) (n=6): 10 µg 10 µL-1 of bupivacaine was administered IT and 0.5 mL of saline was administered intraperitoneally. Group Bupivacaine Ondansetron 1 (Group BO1) (n=6): 10 µg 10 µL-1 of bupivacaine was administered IT and 1 mg kg-1 ondansetron was administered intraperitoneally. Group Bupivacaine Ondansetron 4 (Group BO4) (n=6): 10 µg 10 µL-1 of bupivacaine was administered IT and 4 mg kg-1 ondansetron was administered intraperitoneally. Tail-flick test was applied to rats before and 5,10,20,30,45,60,90,120 minutes after drug administration. Data was showed as the percentage of maximum possible effect (%MPE). Amaç: Bu çalışmanın amacı ratlarda intratekal verilen bupivakainin analjezik etkisine sistemik (intraperitoneal) olarak uygulanan iki farklı doz ondansetronun etkisini araştırmaktır.Gereç ve Yöntem: Araştırmaya yirmi dört adet rat alındı. Grup Serum Fizyolojiğe (Grup SF) (n:6): 10 µL intratekal (‹T) SF+0.5 mL intraperitoneal (‹P) SF, Grup Bupivakain’e (Grup B) (n:6): 10 µg 10 µL-1 ‹T bupivakain + 0,5 mL ‹P SF, Grup Bupivakain Ondansetron 1’e (Grup BO1) (n:6): 10 µg 10 µL-1 ‹T bupivakain + 1 mg kg-1 ‹P ondansetron, Grup Bupivakain Ondansetron 4’e (Grup BO4) (n:6): 10 µg 10 µL-1 ‹T bupivakain + 4 mg kg-1 ‹P ondansetron verildi. ‹laç uygulamalarından önce (0. dk.) ve uygulamalardan sonra 5, 10, 20, 30, 45, 60, 90 ve 120. dk.’larda tail-flick testi uygulandı.
