Effects of defibrotide on blood and aorta malondialdehyde and antioxidant levels in rabbits given atherogenic diet

Abstract

Defibrotide is proposed as an antiatherosclerotic agent in the treatment of cardiovascular diseases. In the present study, the role of free radical-mediated reactions and endogenous antioxidants in the atherosclerotic effect of this drug was assessed. 40 New Zealand type white male rabbits were divided into 4 groups each consisting of 10 animals; Group I (control) was fed with regular chow, GroupII (cholesterol) was given 1% cholesterol in addition to regular chow, Group III (defibrotide) was administered 60 mg/kg/day defibrotide orally and fed with regular chow and Group IV (cholesterol + defibrotide) was given 1% cholesterol and defibrotide was administered the same dose and manner as in Group HI. Before (basal) and after 9 weeks of feeding with each group's special diet regimen, blood cholesterol, malondialdehyde (MDA), catalase (CAT), glutathione (GSH) and glutathione peroxidase (GSH-Px) were determined. In addition to these parameters, glutathione reductase (GSH-Rd) and glutathione S-transferase (GST) were also determined in the aortic tissues. After 9 weeks, blood cholesterol and MDA were elevated and CAT activity was reduced in Group II (cholesterol) compared to the other groups Comparison of the end-experimental values with basal showed that in Group II, the increase in blood cholesterol accompanied a decrease in CAT activity. In Group III (defibrotide), CAT activity was elevated at the end of 9 weeks. Defibrotide receiving groups (III and IV) exhibited an enhanced GSH-Px activity after 9 weeks. Aortic tissue analyses revealed that Group II had the highest tissue MDA concentration. Aortic CAT and GSH-Px activities in Group IV were significantly higher than Group I and III. Morphological examination of aortic tissues exhibited atherosclerotic lesions in Group II and to a lesser extent in Group IV. The results imply that defibrotide might exert antiatherosclerotic effects by oreventins the reduction of antioxidant enzymes in cholesterol- induced atherosclerosis.

Kardiyovasküler hastalıkların sağaltımında defibrotid bir antiaterosklerotik ajan olarak önerilmektedir.Çalışmada, bu ilacın antiaterosklerotik etkisinde, serbest radikal tepkimelerinin ve endojen antioksidanların rolü araştırıldı. 40 adet -Yeni Zelanda tipi beyaz erkek tavşan her biri 10 hayvandan oluşan 4 gruba ayrıldı: Grup I (kontrol) standart diyet ile, Grup II (kolesterol) standart diyet + %1 kolesterol ile, Grup III (defibrotid) standart diyet + defibrotid (60 mg/kg/gün, oral) ile, Grup IV (kolesterol + defibrotid) %1 kolesterol + defibrotid (veriliş şekli ve dozu grup III gibi) ile 9 hafta süre beslendikten önce (bazal) ve sonra kanda kolesterol, malondialdehid (MDA), katalaz (ÇAT), glutatyon (GSH) ve glutatyon peroksidaz (GSH-Px) ölçümleri yapıldı. Bunlara ilave olarak deney sonunda aort dokularında glutatyon redüktaz (GSH-Rd) ve glutatyon S-transferaz (GST) aktiviteleri ölçüldü. 9 haftanın sonunda gruplararası karşılaştırmada, Grup II'de kan kolesterol ve MDA düzeyinin en yüksek, katalaz aktivitesinin ise en düşük olduğu görüldü. Deney sonu değerler bazal ile karşılaştırıldığında Grup II'de kan kolesterolündeki artışa katalaz aktivitesindeki azalmanın eşlik ettiği görüldü. Defibrotid verilen Grup III'te ise 9 haftanın sonunda katalaz aktivitesinin yükseldiği gözlendi. Defibrotid verilen Grup III ve Grup IV'de deney süresinin sonunda GSH-Px aktivitesi de artmıştı. Aort dokusunda en yüksek MDA düzeyleri Grup II'de saptandı. Defibrotid + kolesterol verilen Grup IV'de ise Grup I ve III'e göre artmış katalaz ve GSH-Px aktivitesi gözlendi. Aort dokularının morfolojik incelemesinde Grup II'de daha belirgin, Grup IV'de ise daha az oranda aterosklerotik lezyonlar görüldü. Bu sonuçlar kolesterol ile oluşan aterosklerozda defibrotidin, antioksidan enzimlerin aktivitelerindeki düşüşü engelleyerek antiaterosklerotik etki gösterebileceğini düşündürmektedir.
