Nanoparticulate drug delivery systems for targeting the drugs to the brain

Abstract
A major problem in the treatment of Central Nervous System diseases is the limited entry of the drugs into the brain. The basis for that restricted access is the blood brain barrier, which is formed by the brain capillary endothelium. In recent years, several pharmaceutical strategies have been developed to overcome blood brain barrier. The major of them is a drug targeting to the brain using colloidal drug carrier systems such as nanoparticles. In this review; physiology and structure of blood brain barrier and nanoparticles, its technologies and transport mechanisms to the brain used as brain targeted drug delivery systems were evaluated.
Özet
Santral Sinir Sistemi hastalıklarının tedavisindeki en önemli problem, aktif maddelerin beyne sınırlı geçişleridir. Bu sınırlı geçişin nedeni ise beyin kapiller endoteli tarafından oluşturulan kan beyin engelidir. Son yıllarda, kan beyin engelinin aşılmasına yönelik çeşitli farmasötik stratejiler geliştirilmiştir. Bunlardan en önemlisi, nanopartiküller gibi kolloidal aktif madde taşıyıcı sistemler kullanılarak beyne ilaç hedeflendirilmesidir. Bu derlemede; kan beyin engelinin fizyolojisi ve yapısı ile beyine hedeflenen aktif madde taşıyıcı sistemler olarak kullanılan nanopartiküller, teknolojileri ve beyne geçiş mekanizmaları değerlendirilmiştir.
