Abstract
Background and Purpose: Neuropeptide Y (NPY), one of the most abundant neuropeptides found in the central nervous system (CNS), has been implicated in the regulation of many metabolism. Preproneuropeptide Y (PPNPY) is the signal peptide of the NPY. Effect of PPNPY Leu7Pro polymorphism on metabolic disorder is widely discussed and still controversial. Material and methods: To study the functional aberration in the human wild and mutate types of PPNPY in pathogenesis of type 2 diabetes is hard. Here, the author used a new gene ontology technology to predict the molecular function of human wild and mutate types of PPNPY. Results: Here, it can be seen that there is no functional difference between wild and mutate types of PPNPY. The common functions as Receptor binding, G-protein coupled receptor binding, hormone activity, neuropeptide hormone activity, G-protein coupled receptor activity, calcium channel regulator activity can be seen. Conclusion: This can support the null effect of this polymorphism in clinical findings of the patients. Therefore, this polymorphism might not be an important factor and have null effect in metabolic disorder. Finally, the author hereby recommends for a further study to analyze other know genetic variants within PPNPY gene to add more knowledge.
Özet
Amaç: Santral sinir sistemi (SSS) içinde bol olarak bulunan bir nöropeptid olan Neuropeptide Y (NPY), birçok metabolizmanın düzenlenmesinde ilişiği olduğu gösterilmiştir. Preproneuropeptide Y (PPNPY) NPY’nin sinyal peptididir. PPNPY Leu7Pro polimorfizmin metabolik bozukluklara etkisi geniş olarak tartışılmaktadır ve hala bir sonuca varılamamıştır. Materyal ve yöntem: İnsanda Tip 2 diyabetin patogenezinde PPNPY’nin mutasyona uğramış ve uğramamış olan tiplerinde işlevsel bozuklukların çalışılması zordur. Bu çalışmada yazar, PPNPY’nin insanda mutasyona uğramış va uğramamış olan tiplerinin moleküler fonksiyonunu tahmin etmek üzere yeni bir gen ontoloji tekniği kullanmaktadır.Sonuçlar: Fonksiyonel olarak PPNPY’nin yabani ve mutant tipleri arasında bir fark gözlenmedi. Reseptör bağlama, G-protein çift reseptör bağlama, hormon aktivitesi, nöropeptid hormon aktivitesi, G-protein çift reseptör aktivitesi, kalsiyum kanal düzenleme aktivitesi gibi fonksiyonlar genel işlevler olarak görülebilir. Yargı: Bu durum hastaların klinik bulgularındaki polimorfizmin etkisiz olduğunu destekler. Böylelikle, metabolik bozukluklarda polimorfizmin etkisinin önemsiz olduğu ve önemli bir faktör rolü bulunmadığı söylenebilir. Sonuç olarak, burada yazar, PPNPY geni içindeki diğer bilinen genetik olarak değişik biçimlerin daha fazla bilgiye ulaşmak üzere analiz edilmesi için daha ileri düzeyde bir çalışmayı tavsiye eder.
