Abstract

Glioblastoma (GBM) is an aggressive primary brain tumor and it is known that prognosis ispoor. Because of the infiltration of the tumor cells to the whole brain tissue, surgery is donefor decompression and for histopathological diagnosis. Radiotherapy is mandatory aftersurgery but survival period is not sufficient. In recent years there are many researches forchemotherapeutical agents to use for treatment of GBM but most of the results of theresearches are controversial. Temozolomide (TMZ) is a new agent which is effective inprolonging survival of GBM patients. Erythropoietin promotes red blood cell maturation andused to prevent or treat anemia. It is also known that EPO has neuroprotective effect. In thisstudy we used GMS-10 human glioblastoma cell lines. We applied TMZ with different dosesand TMZ + EPO combination to check the efficacy on the survivin gene expression in humanglioblastoma cell lines. As a result we observed that increasing the dose of TMZ andcombination with EPO results over expression of survivin gene which means drug resistance.

Glioblastoma (GBM) çok agresif bir beyin tümörüdür ve prognozu kötüdür. Tümörhücrelerinin beyin dokusuna yaygın infiltrasyonu nedeniyle cerrahi girişim sadecedekompresyon ve doku tanısı amacıyla uygulanabilir. Cerrahi sonrası radyoterapi mutlakgereklidir ancak ortalama yaşam süresi yeterli değildir. Son yıllarda GBM tedavisindekullanılmak üzere kemoterapötik ilaç araştırmaları yoğunlaşmıştır ancak çoğu çalışmadasonuçlar tartışma konusu olmaktadır. Temozolomide (TMZ) GBM hastalarında yaşamsüresini uzatabilen yeni bir ajandır. Eritropietin (EPO) ise anemi korunmasında veyatedavisinde kullanılan bir ajan olmakla beraber nöroprotektif özelliği de bilinmektedir. GMS- 10 GBM hücre kültürlerinde TMZ' nin iki farklı doz ve Eritropoietin kombinasyonu ilesurvivin geni üzerindeki etkilerini araştırdık. Sonuçta TMZ dozu artırılmasının ve EPOkombinasyonunun survivin geni ekspresyonunu artırdığı, yani ilaç direncine neden olduğunusaptadık.
