Kortikal Displazi Oluşturulmuş Sıçanlarda Melatonin ile Kannabinoid Reseptörü İlişkisi

Abstract
Objectives: Cortical dysplasia is a developmental malformation that occurs as a result ofdefects in neuronal proliferation, migration and organization during the formation of braincortex layers. Cannabinoid-1 Receptor (CB1R) plays a role in brain cortex development.Melatonin neuroprotective is an agent. In this study using an experimental cortical dysplasiamodel, CB1R expression and the effect of melatonin on CB1R were investigated.Methods: Eighteen female Wistar albino rats were randomly divided into three groups (n=6).A single dose of intraperitoneal (i.p) saline (20 ml/kg) was administered in the 15th day ofpregnancy in the rats group 1 (sham group). A single dose of i.p BCNU (20 mg/kg) wasadministered in the 15th day of pregnancy in the rats in group 2 (BCNU) and group 3(BCNU+ Melatonin). In addition, i.p melatonin (10 mg/kg) starting on day 15 of pregnancyand continuing until the day of delivery were administered to the rats in group 3(BCNU+Melatonin). Newborn brain sections were investigated using Luxol fast blue andanti-CB1R staining.Results: While the sham group had cerebral cortex stratification compatible with the normalnewborn histology, it was observed that the stratification was disrupted in the group 2 andgroup 3. In the observed brain regions (cerebral cortex, cerebellar cortex and hippocampus), itwas observed that CB1R immunoreactivity decreased in group 2 and 3.Conclusion: In an experimental cortical dysplasia model, it was seen that the administrationof melatonin revealed a protective effect against the decrease in CB1R expression. It has beensuggested that a decrease in CB1R expression would also play a role in cortical dysplasiaformation mechanisms.

Amaç: Kortikal displazi beyin korteks tabakalarını oluşturan nöronların proliferasyonu,migrasyonu ve organizasyonunun bozulması sonucu oluşan gelişimsel bir malformasyondur.Kannabinoid-1 Reseptör (CB1R) beyin korteks gelişiminde rol oynar. Melatoninnöroprotektif etkileri olan bir ajandır. Bu çalışmada deneysel kortikal displazi modelioluşturularak CB1R ekspresyonu ve melatoninin CB1R üzerine etkisi etkisi incelendi.Method: 18 dişi Wistar albino sıçan rasgele 3 gruba ayrıldı (n=6). Grup 1 (Sham grup)gebeliğin 15. gününde intraperitoneal (i.p) tek doz 20 ml/kg serum fizyolojik uygulandı. Grup2 (BCNU) ve Grup 3 (BCNU+Melatonin) gebeliğin 15. gününde i.p tek doz BCNU (20mg/kg) uygulandı. Grup 3 (BCNU+Melatonin) de, BCNU uygulamasına ek olarak gebeliğin15. gününde başlayarak doğuma kadar i.p melatonin (10 mg/ kg) uygulandı. Yenidoğan beyinkesitleri Luksol fast blue ve CB1R antikoru ile boyanarak incelendi.Sonuç: Sham grubu, normal yenidoğan serebral korteks tabakalanması ile uyumlu iken, grup1 ve grup 2 de tabakalanmanın bozulduğu görüldü. İncelenen beyin bölgelerinde (serebralkorteks, serbellar korteks, hipokampus) CB1R boyanması, grup 2 ve grup 3 de azalmıştı.Tartışma: Deneysel kortikal displazi modelinde uygulanan melatoninin CB1Rekspresyonundaki azalmaya karşı koruyucu etkisi olduğunu gözlemledik. Kortikal displazioluşum mekanizmaları içinde CB1R ekpresyonundaki azalmanında rolü olabileceğiöngörülmektedir.
