Effect of sodium lactate on the growth kinetics of Bacillus cereus

Abstract
Models describing the lag phase duration (LAG) and growth rate (GR) of Bacillus cereus as a function of temperature, pH, sodium lactate and sodium chloride concentrations were obtained in this study. Gompertz function was used to calculate the LAG and GR of B. cereus. These parameters were expressed as a function of the independent variables using the polynomial models. In order to obtain a residual distribution closer to a normal distribution, the natural logarithm of the growth kinetic parameters were used in modeling. For reasons of parsimony, the polynomial models were reduced to contain only the coefficients significant at a level of $pleq 0.05$. Using the reduced polynomial models ($R^2$ of 95.2 and 93.9 for In LAG and In GR models, respectively), three dimensional response surface graphs were drawn illustrating the effects of the independent variables on the growth parameters. Results of the modeling study indicated that all of the studied factors had a significant affect on the growth of B. cereus in the experimental range studied. The antimicrobial activity of sodium lactate was found to be strongly affected by the pH ($pleq 0.05$) and relatively to a lesser extent by temperature ($pleq 0.05$). Antimicrobial activity involved both prolonged lag phase duration and reduced growth rate. No effect of sodium chloride was found on the antimicrobial activity of sodium lactate.

Bu çalışmada, sıcaklık, pH, sodyum laktat ve sodyum klorür konsantrasyonlarının Bacillus cereus’un lag fazı süresi (LAG) ve çoğalma hızı(GR) üzerindeki etkisini ifade eden matematiksel modeller oluşturulmuştur. B. cereus için LAG ve GR değerlerini hesaplamak amacıyla Gompertz fonksiyonu kullanılmıştır. Bu kinetik parametreler, polinomal model kullanılarak bağımsız değişkenlere bağlı olarak ifade edilmiştir ve bağımsız değişkenlerin çoğalma parametreleri üzerindeki etkilerini gösteren üç boyutlu grafikler çizilmiştir. Çalışmanın sonuçları, sodyum laktatın antimikrobiyal aktivitesinin büyük oranda pH’dan ($pleq 0.05$), daha düşük oranda ise sıcaklıktan ($pleq 0.05$) etkilendiğini göstermiştir. Sodyum klorürün, sodyum laktatın etkinliği üzerinde bir etkisi bulunmamıştır.
