Survival of Listeria monocytogenes in İnegöl meatballs refrigerated at 4 degree

Abstract

This study was performed in order to determine the survival of Listeria monocytogenes refrigerated at 4°C in İnegöl meatballs consisting of veal and lamb's meat and has a high pH value. For this purpose, 1.3x $10^3$ cfu/g, 3.2x $10^3$ cfu/g, 1.0x $10^4$ cfu/g and 1.4x $10^5$ cfu/g of Listeria monocytogenes serotype 4b was inoculated into four groups of İnegöl meatballs. In these İnegöl meatballs refrigerated at 4°C, Listeria monocytogenes counts were determined on days 0, 1, 2, 3, 4, 5, 6 and day 7. Also total mesophyll aerobic bacteria, psycrophilic bacteria, coliform bacteria, lactobacilli, staphylococci and micrococci, yeast and mold, sulphate reducing anaerobs, koagulase pozitive staphylococci, and E. coli counts were determined. And also pH values were determined during the storage.During refrigeration, no significant changes or decreases were observed in the Listeria monocytogenes counts in all four of the inoculation levels, and these values remained nearly the same. In the end of the refrigeration, the first, the second, the third and the fourth inoculation values were determined as 1.9x $10^3$ cfu/g, 8.1x $10^3$ cfu/g, 2.0x $10^4$ cfu/g and 3.7x $10^5$ cfu/g, respectively. As a result, the wide presence of Listeria monocytogenes in ground meat, the high possibility of cross contamination in meatballs, the survival of this pathogen and keeping its numbers in the same levels even if meatballs are stored under refrigeration indicates that İnegöl meatballs can be a potential public hazard. Therefore, the importance of through grilling of İnegöl meatballs is an unavoidable step.

Özet

Bu çalışma, dana ve kuzu eti karışımından elde edilen ve yüksek pH değerine sahip olan İnegöl köftelerin buzdolabında saklanması sırasında Listeria monocytogenes'in yaşamım incelemek amacıyla yapıldı. Bu amaçla, dört gruba ayrılan İnegöl köftelere 1.3x $10^3$ kob/g, 3.2x $10^3$ kob/g ve 1.0x $10^4$ kob/g ve 1.4x $10^5$ kob/g düzeyinde L. monocytogenes serotip 4b inokule edildi. 4°C'de buzdolabında muhafazaya alınan İnegöl köftelerde, muhafazanın 0., 1., 2., 3., 4., 5., 6., 7., günlerinde L. monocytogenes sayıları belirlendi ve toplam mezofıl aerob, psikrofil bakteriler, koliform bakteriler, laktobasiller, stafilokok ve mikrokoklar, maya ve küfler, sülfit indirgeyen anaeroblar, koagülaz pozitif stafilokoklar, E.coli bakımından analizlere tabi tutuldu. Ayrıca İnegöl köftelerin muhafaza süresi içinde pH tayini yapıldı. Dört inokulasyon düzeyinde de Listeria monocytogenes sayısında önemli bir değişiklik olmadı ve İnegöl köftelerdeki Listeria monocytogenes sayıları korundu. Muhafaza süresinin sonunda Listeria monocytogenes sayıları 1. inokulasyon düzeyinde 1.9x $10^3$ kob/g, 2. inokulasyon düzeyinde 8.1x $10^3$ kob/g, 3. inokulasyon düzeyinde 2.0x $10^4$ kob/g ve 4. inokulasyon düzeyinde de 3.7x $10^5$ olarak tespit edildi. Sonuçta, kıymalarda Listeria monocytogenes'in yaygın olarak bulunmasına bağlı olarak, köftelerde de kontaminasyon olasılığının yüksek olduğu ve köftelerin buzdolabında muhafazası sırasında da etkenin aynı düzeylerde varlığını devam ettirmesi İnegöl köftenin halk sağlığı için potansiyel bir tehlike olabileceğini göstermektedir. Böylece tüketilmeden önce İnegöl köftelerin etkili bir pişirme işlemine tabi tutulmasının önemi ortaya çıkmaktadır.
