Effects of aluminum toxicity and pentylenetetrazol-induced epileptic seizures on blood-brain barrier

Abstract
We studied the effects of aluminum toxicity and pentylenetetrazole-induced status epilepticus during this aluminum toxicity on the permeability of blood-brain barrier (BBB). Rats were injected with aluminum-L-glutamate (Al-L-glu), L-glutamate (L-glu) and physiological saline subcutenously. Following the administration of these substances for twelve weeks, Evans blue (EB) and horseradish peroxidase (HRP) tracers were used to evaluate the functional state of BBB. Epileptic seizures were induced by intravenous injection of pentylenetetrazole (PTZ). The alterations of the permeability for EB and HRP were examined from four different brain regions. The results indicated that EB extravasation was substantially increased in all brain regions of animals in aluminum, aluminum plus PTZ, and PTZ groups. However, the glutamate group showed no BBB permeability change as well as control group. The increase in extravasation of EB into cerebral tissue as a result of aluminum plus PTZ was markedly higher than EB values that obtained in both by aluminum and PTZ groups. It was also demonstrated that HRP was both penetrated to endothelial cytoplasm and extracelluler portion of the brain in the same groups excluded from glutamate and control groups. These finding indicate that endothelial cells were affected in the prolonged (chronic) exposure to aluminum and that these affected cells could more easily respond to pentylenetetrazole-induced status epilepticus as a secondary effect.

Alüminyum intoksikasyonu ve bu intoksikasyon sırasında oluşturulan konvulsiyonların kan-beyin bariyer (K-BB) permeabilitesine olan etkileri çalışıldı. Alüminyum-L-glutamat (Al-L-glu), L-glutamat (L-glu) ve serum fizyolojik sıçanların deri altına enjekte edildi. Bu maddelerin oniki hafta süre ile verilmesini takiben, Evans blue (EB) ve horseradish peroksidase (HRP) traserleri K-BB' nin fonksiyonel durumunu değerlendirmek için kullanıldı. Epileptik nöbetler pentilentetrazolün (PTZ) intravenöz enjeksiyonuyla oluşturuldu. Uygulanan traserlerle ilgili K-BB permeabilite değişiklikleri beynin dört farklı bölgesinde incelendi. Alüminyum, PTZ ve alüminyum+PTZ gruplarında incelenen beyin bölgelerine EB boya geçişinin önemli oranda arttığı gösterildi. Fakat, kontrol grubunda olduğu gibi, glutamat grubunda da beyine EB geçişi gösterilemedi. Alüminyum+PTZ grubunda beyin dokusuna geçen EB miktarındaki artış, sadece alüminyum veya PTZ grubunda elde edilen EB değerlerinden çok yüksekti. Ultrastrüktürel düzeyde de kontrol ve glutamat grupları hariç olmak üzere alüminyum, aluminyum+PTZ ve PTZ gruplarının incelenen beyin bölgelerinde HRP' nin hem endotel sitoplazmasına hem de beyin ekstrasellüler tarafına geçtiği gösterildi. Bu bulgular, kronik alüminyum intoksisitesinde endotel hücrelerinin etkilendiğini ve etkilenen bu hücrelerin ise sekonder etki olarak uygulanan nöbetlere karşı daha kolay cevap verebildiğini göstermektedir.
