The role of Kupffer cell inactivation in D-galact-osamine induced hepatocellular damage in rats

Abstract
In the present study we examined the role of Kupffer cells in galactosamine (Gal-NH2) induced hepatitis and also the capability of gadalinium chloride (GdCl3) -a specific Kupffer cell inhibitor-to prevent the liver damage in rats. Following acute Gal-NH2 (1g/kg) injection to the rats, plasma ALT, AST, GGT activities and liver lipid peroxide levels were increased significantly and the liver GPx, GST and SOD activities were not changed in comparison to the controls. GdCl3 (10mg/kg) administration , In addition to Gal-NH2 did not improve the increased plasma ALT, AST, GGT activities and lipid peroxide levels in 48.hours in the rats. In conclusion, we thought that Kupffer cell activation does not take role in Gal-NH2 induced hepatic injury.
Özet
Çalışmamızda, galaktozamin (Gal-NH2) uygulanarak hepatosellüler hasar oluşturulan sıçanlarda spesifik Kupffer hücresi inaktivatörü olan gadalinyum klorür'ün (GdCl3) hasarın önlenmesindeki etkilerini inceledik. Akut Gal-NH2 (1g/kg) enjeksiyonunu takiben sıçanların plazma ALT, AST, GGT aktiviteleri ile karaciğer lipit peroksit düzeylerinde anlamlı artışlar saptanmıştır. Gal-NH2 uygulaması sıçanlarda karaciğer GPx ve SOD aktivitelerinde kontrollere göre anlamlı bir değişikliğe neden olmamıştır. Gal-NH2 uygulanarak hepatoselluler hasar oluşturulan sıçanlara GdCl3 (10 mg/kg) verilmesi 48. saate artmış plazma ALT, AST, GGT aktiviteleri ile karaciğer lipit peroksit düzeylerinde bir düzelme sağlamamıştır. Elde ettiğimiz sonuçlar bize, Gal-NH2'nin hepatotoksik etkisinde Kupffer hücre aktivasyonunun rolü olmadığını düşündürdü.
