The value of the assessment of CD 55 and CD 59 in the peripheral blood monocytes and granulocytes in patients with aplastic anemia: A study in 20 cases

Abstract
A study in 20 cases. Peripheral blood monocytes and granulocytes of 14 healthy persons and 12 cases of severe and 8 cases of very severe aplastic anemia were investigated with flow cytometry for CD 55 and CD 59 expressions. Ham and sugar-water tests were also performed in the patient group. The patients could be divided into two subgroups according to the level of expression of CD 55 and CD 59. Thus, in 9 patients the expression of CD 55 and CD 59 were within normal limits whereas in 11 patients there expressions were less than the lower limits of normal values. In none of the twenty patients, the Ham and the sugar-water tests were found positive during the mean follow up period of 22 months (range 3-57 mo). It has been observed that responses of the patients to treatment (antilymphocyter globulin, cyclosporine A, granulocyte colony stimulating factor, methylprednisolone) were parallel to their values of expressions namely those with low CD 55 and CD 59 expressions responded poorly to the immunsuppressive treatment than those with normal values.

Sağlıklı 14 şahısta ve 8'i çok ağır, 12'si ağır aplastik anemili vakalarda tedaviye başlamadan evvel çevre kanı monosit ve granülositlerinde "flow" sitometreyle CD 55 ve CD 59 ifadeleri incelendi. Bunlara ilaveten hasta grubuna kronik hemoliz testlerinden Ham ve şeker-su testi yapıldı. Hastalar, her bir hastada çevre kanı monosit ve granülositlerinde tespit edilen CD 55 ve CD 59 ifadelerinin normal gruptaki herhangi bîr değerin minimumundan küçük oluşuna, göre iki gruba ayrıldı. Böylece 9 hasta CD 55 ve CD 59 ifadeleri normal sınırlarda ve 11 hastada ise bunların ifadeleri normalden düşük olarak tespit edildi. Yirmi hastanın hiçbirinde takip edildikleri ortalama 22 (3-57) ay içinde Ham ve şeker-su testleri pozitif bulunmadı. Antilenfositer globulin, siklosporin A, granülosit koloni uyarıcı faktör ve metil prednizolondan oluşan yoğun immunsupressif tedaviye CD 55 ve CD 59 ifadeleri düşük olan hastaların cevaplarının normal bulunanlardan kötü olduğu tespit edildi.
