Line immunoblot confirmation of third generation elisa in the diagnosis of hepatitis C virus ( HCV ) infection

Abstract

Diagnosis of HCV infection can be done by detection of viremia or antibodies to viral proteins. Supplementary assays are recommended to confirm positive ELISA anti-HCV results of asemptomatic cases with normal ALT. In this study, the sera of 37 anti-HCV positive (ELISA III) cases by Third Generation HCV Line Immunoblot (LIATEK III). Twenty-one cases were positive, five cases were indeterminate and 11 cases were negative. Cases with elevated ALT level were more in the line Immunoblot positive group. All the cases with negative line Immunoblot had not only normal ALT, but low level positivity with ELISA as well.

Özet

Hepatit C virus (HCV) infeksiyonunda tanı, virusa karşı gelişmiş antikorların ya da doğrudan vireminin gösterilmesiyle olanaklıdır. Antikor testlerinden ELISA ile pozitif bulunan özellikle asemptomatik ve ALT değerleri normal kişilerin destekleyici testler ile doğrulanması önerilmektedir. Bu çalışmada, ELISA III ile anti-HCV pozitif olan 37 hasta serumunda doğrulama testi olarak üçüncü generasyon HCV Line Immunoblot (LIATEK III) kullanıldı. Yirmi-bir hasta pozitif, beş hasta indeterminate, 11 hasta negatif olarak değerlendirildi. Line Immunoblot pozitif grupta ALT yüksek hasta sayısının daha fazla olduğu saptandı. Line Immunoblot negatif hastaların hiçbirinde ALT yüksekliği bulunmadığı gibi ELISA ile de düşük titre pozitiflik olduğu görüldü.
