Gas chromatographic analysis in microbial identification: A study on Legionella pneumophila

Abstract

Gas chromatography of bacterial cellular fatty acid methyl esters (FAME) is used in clinical microbiology for identification of many medically important bacteria. Total fatty acid composition of a microorganism is an important taxonomic character and that fatty acid data can be analyzed quantitatively to provide useful taxonomic information at the species level and in some cases, the subspecies level. The reproducibility of the chromatographic technique is determined by repeated analyses of a standard quantitative FAME mixture (Microbial ID Inc., Newark, Del.) (MIDI). In this study, seven Legionella pneumophila strains that were isolated from 113 water samples were confirmed by gas chromatography method in addition to other confirmation methods.

Özet

Bakteriyel hücresel yağ asit metil esterlerinin (YAME) gaz kromatografisi klinik mikrobiyolojide medikal olarak önemi olan birçok bakterinin identifikasyonu için kullanılmaktadır. Bir mikroorganizmanın toplam yağ asit bileşimi önemli bir taksonomik özelliktir ve bu bilgi kantitatif olarak analiz edilerek bazı olgularda alt tür düzeyinde olmak üzere, tür düzeyinde kullanışlı taksonomik bilgi sağlar. Kromatografik tekniğin tekrarlanabilirliği standart bir kantitatif YAME karışımının yineleyen analizi ile sağlanır (Microbial ID Inc., Newark, Del.) (MIDI). Bu çalışmada 113 su örneğinden izole edilen yedi Legionella pneumophila suşunun, diğer doğrulama yöntemlerinin yanı sıra, gaz kromatografisiyle doğrulaması gerçekleştirilmiştir.
