The comparison of conventional method with automated systems for the identification of gram-negative bacteria

Abstract

In our day, in addition to the widespread use of conventional method for bacteriological identification, the laboratories also have the chance of relatively more rapid reporting by using automatized identification system which has been developed by modifying the conventional method. This study aimed at comparing automated systems with conventional method in the identification of Gram-negative bacteria. In the study, 271 Gram-negative bacteria belonging to Family Enterobacteriaceae and 75 non-fermentative Gram-negative bacteria (totally 346) were evaluated using A TB ID 32 GN and A TB Rapid ID 32 E strips (API system)and conventional identification methods. According to the identification results of enteric bacteria using conventional method and API system, the compatibility was found to be 93.0 % and 91.1 %, for genus and species level, respectively. The compatibility was found to be 84.0 % and 74.7 %, for genus and species level, respectively, for non-fermentative Gram-negative bacteria. One point one percent of enteric bacteria remained unidentified using API system and the conventional method. For the non-fermentative bacteria the unidentification rate was 4.0 % for the conventional method and 2.7 % for the API system. Conventional method was found to be cheaper than API system when cost was analyzed.

Bakteri identifikasyonunda konvansiyonel yöntemler hala çok yaygın olarak kullanılmakla beraber, bu yöntemlerin modifiye edilmesiyle geliştirilen otomatize identifikasyon sistemleri sayesinde laboratuvarlar daha kısa sürede sonuç verme şansını yakalamışlardır. Bu araştırmada Gram-negatif bakteri identifikasyonunda konvansiyel ve otomatize sistemlerin karşılaştırılması amaçlanmıştır. Çalışmada, 271'i Enterobacteriaceae familyasına, 75'i non-fermentatif Gram-negatif bakterilere ait olmak üzere toplam 346 bakteri ATB İD 32 GN ve ATB Rapid ID 32 E stripleri kullanılarak Analytical Profile Index (API) sistemi ve konvansiyonel identifikasyon yöntemi ile değerlendirilmiştir. Enterobacteriaceae'ların iki sistemle identifikasyon sonuçlarına göre uyum yüzdesi cins düzeyinde % 93.0, tür düzeyinde % 91.1 olarak bulunmuştur. Non-fermentatif Gram-negatif bakteriler arasındaki uyum cins düzeyinde % 84.0, tür düzeyinde % 74.7 olarak saptanmıştır. Enterobacteriaceae'nin konvansiyonel yöntemle ve API sistemi ile % 1.1'i, non-fermentatif bakterilerin ise konvansiyonel yöntemle % 4.0'ü, API sistemiyle % 2.7'sinin doğru olarak tanımlanması mümkün olmamıştır. Maliyet analizi sonuçlarına göre konvansiyonel yöntem ile identifikasyonun daha ucuz olduğu saptanmıştır.
