In vitro pexiganan susceptibility of clinical isolates of aerobic bacteria

Abstract
Pexiganan, an antimicrobial peptide, is a synthetic analog of the magainin II peptides originally isolated from the skin of the African clawed frog. In this study, pexiganan was shown to have broad-spectrum antimicrobial activity when it was tested against 580 clinical isolates of Gram-positive and Gram-negative aerobic bacteria by broth microdilution method as recommended by NCCLS. The MIC90 values of pexiganan were detected as 32 $mu$g/ml for Escherichia coli, Klebsiella spp., Enterobacter spp. and 64 $mu$ g/ml for Pseudomonas aeruginosa. The MIC90 values of pexiganan for Staphylococcus aureus, coagulase-negative staphylococci, and Enterococcus faecium were 16 $mu$g/ml. Enterococcus faecalis and Proteus spp. were found to be less susceptible to pexiganan with both MIC50 and MIC90 values greater than 256 $mu$g/ml These results demostrate that pexiganan could serve as a potent topical antimicrobial substance.

Antimikrobiyal bir peptit olan peksiganan orjinal olarak Afrika kurbağasının derisinden elde edilmiş magainin II grubu peptitlerin sentetik analoğudur. Bu çalışmada, peksiganan Gram-pozitif ve Gram-negatif 580 klinik kökende NCCLS önerileri doğrultusunda mikrodilüsyon yöntemiyle denendiğinde, geniş spektrumlu antimikrobiyal aktiviteye sahip olduğu gösterilmiştir. Peksigananın MİK90 değerleri Escherichia coli, Klebsiella spp., Enterobacter spp. için 32 $mu$g/ml ve Pseudomonas aeruginosa için 64 $mu$g/ml olarak saptandı. Staphylococcus aureus, koagülaz-negatif stafilokoklar, Enterococcus faecium için peksigananın MİK90 değeri 16 $mu$g/ml idi. Enterococcus faecalis ve Proteus spp. MIC50 ve MIC90 256 $mu$g/ml'den daha yüksek değerleriyle peksiganana az duyarlı bulundu. Bu sonuçlar, peksigananın etkili bir topikal antimikrobiyal madde olabileceğini göstermektedir.
