Abstract

The purpose of the study was to determine the optimum detection time for Brucella melitensis by BACTEC 9120 (Becton Dickinson Diagnostic Instrument Systems) blood culture system. The study was performed with 12 B. melitensis strains isolated from blood cultures. A suspension of each strain was made in 5 ml of trypticase soy broth (TSB) and adjusted to approximately McFarland No. 1 standard. These suspensions consisting of 3x$10^8$ cfu/ml were then diluted serially with TSB to 3x$10^2$ cfu/ml. Each BACTEC 9120 aerobic bottle was inoculated with 5 ml of fresh blood collected from healthy volunteers. Dublicate bottles were then inoculated with 0.05 ml and 0.5 ml (15 cfu/ml and 150 cfu/ml) of 3x$10^2$ cfu/ml dilution. All bottles were transiently vented, inserted into the instrument, and monitored until they became positive. Bu çalışmanın amacı, BACTEC 9120 (Becton Dickinson Diagnostic Instrument Systems) otomatik kan kültür sisteminin Brucella melitensis'i saptama süresinin bakteri konsantrasyonu ile ilişkisini saptamak idi. Bu amaçla, kan kültürlerinden izole edilen 12 B. melitensis suşu çalışma kapsamına alındı. Her bir suşun 5 ml triptikaz soy buyonda McFarland 1'e göre 3x$10^8$ koloni içeren süspansiyonu hazırlandı. Seri sulandırımlarla 3x$10^2$'lik süspansiyon elde edilerek buradan alınan 0.05 ve 0.5 ml(15 cfu/ml ve 150 cfu/ml) örnekler, sağlıklı gönüllülerden alman 5'er ml taze insan kanı ilave edilmiş olan BACTEC 9120 aerop kan kültürü şişelerine ekildi.
