Protease inhibitor system polymorphism in horses

Abstract

A total of 85 Arabian and 104 Throughbred horse were examined in respect to protease inhibitor phenotypes by acid gradient polyacrylamide gel electrophoresis (PAGE). A combination of protein, antitrypsin and antichymotrypsin staining were performed after gradient polyacrylamide gel electrophoresis. , Thirtyfive phenotypes arising from twelve allels namely $Pİ^F, Pİ^G, Pi^I ,Pİ^L, Pİ^{L2}, Pİ^{L*},Pİ^N, Pi^P, Pi^S, Pİ^T, Pi^U, Pi^Z$ were recognized in the course of this study .The gene frequencies of the allels were calculated. Gene frequencies of variant $Pi^L$ (0.418) in Throughbreds and $Pi^U$ (0.357) in Arab horses were discovered to be the highest. $Pi^P$ and $Pi^Z$ in Throughbreds, $Pi^{L*}, Pi^N, Pi^T$ in Arabs were not observed in this population.

Bu çalışmada poliakrilamid jel elektroforezi (PAGE) yöntemi ile 85 Safkan Arap ve 104 Safkan İngiliz atında proteaz inhibitor (Pi) sistemine ait fenotipler belirlenmiştir. Elektroforez işleminden sonra jeller protein ve antitirpsin, antikemotiripsin boyama metodu ile boyanmıştır. $Pİ^F, Pİ^G, Pi^I ,Pİ^L, Pİ^{L2}, Pİ^{L*},Pİ^N, Pi^P, Pi^S, Pİ^T, Pi^U, Pi^Z$ olmak üzere 12 allel ile kontrol edilen 35 fenotip saptanmış ve gen frekansları hesaplanmıştır. Allel gen frekansı Safkan İngiliz at ırkında $Pi^L$ (0.418),Safkan Arap atlarında ise $Pi^U$ (0.357) allelerinde en yüksek olarak saptanmıştır. Safkan Arap atlarında $Pi^{L*}, Pi^N, Pi^T$, Safkan İngilizlerde ise $Pi^P$ ve $Pi^Z$ ailelerine rastlanmamıştır.
