Özet

Bu çalışmada; Sequoia sempervirens (Lamb.) (Sahil Sekoyası) Endi. 'in hızlı gelişme gösteren üstün fertlerinden oluşan 6 farklı ortetten alınan sürgün uçlarının in-vitro koşullar altında indol bütirik asil (IBA) ve indol asetik asit (IA A) farklı dozlarının plantet gelişimi üzerine etkisi incelenmiştir. Denemelerde Murashige&Skoog MS besiyeri kullanılmıştır. MS besiyerinde 1AA (2, 1.5, l, 0.5 mg/L ) ve IBA (4, 3.5, 3, 2.5 mg/L)'mn dörder farklı konsantrasyonunun planletin sürgün ve kök oluşumuna etkileri araştırılmıştır. Ayrıca, planletlerin 7 farklı yetişme ortamındaki adaptasyon yetenekleri belirlenmiştir. Sonuç olarak, farklı ortetlere ait explantlarin in-vitro koşullarındaki gelişimlerinin farklı olduğu; besiyerlerdeki 1AA dozu azaldıkça ve IBA dozu arttıkça planletin kök/gövde gelişiminde olumlu değişimler olduğu tespit edilmiştir. Farklı yetişme ortamlarından ise % 70 Barma turbası + % 30 kompost çay artığı ortamı en iyi sonucu vermiştir.

Abstract

In this study; six ortets were selected from fast growing plus trees of Sequoia sempervirens (Lamb.) Endl. in which shoot tips are used as the propagation materials. The effects of different concentrations ofindole butyric acid (IBA) and indole acetic acid (IAA) were examined for planlets growing under in-vitro conditions. Murashige&Skoog medium MS was used as a four different concentrations oflAA (2, 1.5, 1, 0.5 mg/L) and IBA (4,3.5, 3, 2.5 mg/L) in order to examine the effect of these concentrations on the root and shoot developing characteristics. In addition, adaptation of planlets was studied on seven different growth médias. As a result, different ortets showed significant differences in-vitro condition as an decreasing IAA and increasing IBA concentrations effected to the possitive growth of root and shoot development of planlets. The best growth media was found to be 70% Barma peat + 30% tea compost.
