Cryopreservation of Kangal Dog Semen in Commercial Extender (CaniPlus Freeze)

Abstract

Various solutions are used as sperm extenders in order to keep dog spermatozoa for a short time or for freezing for a long time. In our study, it was aimed to determine the effect of commercial diluent on freezing-thawing spermatological parameters in freezing of Kangal dog sperm. For this purpose, a total of 32 ejaculates were taken from two Kangal dogs by massage method. After freezing, the semen was divided into two equal portions and the first portion was diluted with Tris solution and the second portion was diluted with CaniPlus Freeze diluent. spermatozoa were placed in the refrigerator (+4 ⁰ C) and left for 1.5 hours. At the end of the equilibration period, the straws were frozen for 20 minutes at a height of 4 cm (-120 ⁰ C) from the level of liquid nitrogen and stored therein by immersion into liquid nitrogen. After freezing, the semens were thawed at 38 ⁰ C for 30 seconds and subjected to spermatological examination. In the study, the resulting motility ratios were 45% and 50%, respectively in the control and CaniPlus Freeze groups (P>0.05). Abnormal spermatozoon ratio was found to be 18.8% in control group and 16.4% in CaniPlus Freeze group (P>0.05). As a result, in addition to researchers, we are particularly considering that the use of commercial diluents by clinician veterinarians in dogs will be of practical benefit in semen storage studies.

Günümüzde köpek spermasının kısa süreli ya da dondurularak uzun süreli saklanması amacıylasperma sulandırıcısı olarak çeşitli solüsyonlar kullanılmaktadır. Bu çalışmada Kangal köpeğispermasının CaniPlus Freeze sulandırıcısı kullanılarak dondurulabilirliğinin araştırılmasıamaçlanmıştır. Bu amaçla ırka ait fenotipik özellikleri taşıyan iki adet Kangal köpeğinden masajyöntemi ile alınan toplam 32 ejekülat sperma kullanılmıştır. Dondurma işlemi için birleştirme(pooling) sonrası sperma iki eşit kısma bölünerek birinci kısım Tris solüsyonu ile ikinci kısım iseCaniPlus Freeze sulandırıcısı ile sulandırılmıştır. Payetlere çekilen spermalar buzdolabına (+4 ⁰ C)yerleştirilmiş ve 1.5 saat ekilibrasyona bırakılmıştır. Ekilibrasyon süresi sonunda payetler, sıvı azotseviyesinden 4 cm yükseklikte (-120 ⁰ C) 20 dakikada dondurulmuş ve buradan sıvı azotadaldırılarak saklanmıştır. Dondurma işleminden sonra spermaların bulunduğu payetler 38 ⁰ C’dekisu banyosunda 30 saniyede çözdürülmüş ve spermatolojik muayeneye tabi tutulmuştur. Çalışmadaçözüm sonu motilite oranları kontrol ve CaniPlus Freeze grubunda sırasıyla %45 ve %50 olarakbelirlenmiştir (P>0.05). Anormal spermatozoon oranı ise kontrol grubunda %18.8 iken CaniPlusFreeze grubunda %16.4 olarak tespit edilmiştir (P>0.05). Sonuç olarak araştırmacıların yanı sıraözellikle klinisyen veteriner hekimlerin köpeklerde ticari sulandırıcıları kullanmasının spermasaklama çalışmalarında pratik fayda sağlayacağı kanısına varılmıştır.
