Abstract

Objective: Due to the incidence and high death rate, cancer is one of the important health problems together with environmental carcinogenic factors today. Cancer is most frequently seen in females. Chemotherapeutic agents such as cyclophosphamide and doxorubicin were administered for the cancers seen in females. Previous studies indicated that these chemotherapeutic agents have an evident effect in germ cells. Vitamin D prevents apoptosis of cells by both inhibiting angiogenesis and proliferative effect. The effect of vitamin D on the damage of ovarium tissue by Cyclophosphamide and doxorubicin is not known yet. In this study, it was aimed to investigate the effects of vitamin D on the changes on rat ovarium tissue and it's function by cyclophosphamide and doxorubicin administration. Materials and Methods: Thirty-six female rats were used in the study. The rats were divided into six groups including 6 rats each: control group, doxorubicin group, doxorubicin plus vitamin D group, cyclophosphamide group, cyclophosphamide plus vitamin D group and vitamin D group. All rats were decapitated under anesthesia after the experiments. Ovarium tissues were immediately extracted and histopathologic sections were prepared and ovarium reserves were examined. Results: Whereas ovarium reserve was found as sufficient in control and vitamin D groups, decrease was observed in the ovarium reserves of the other groups. The maximum decrease was seen in cyclophosphamide group. Conclusion: As a result it was seen that substantial dose of vitamine D had no preventive effect on cytotoxic effect of cyclophosphamide and doxorubicin

Özet

Amaç: Kanser hastalığının, sık görülmesi ve yüksek oranda ölümlere yol açması, artan çevresel kanser yapıcı etkenler nedeni ile günümüzün önemli sağlık sorunlarından birisidir. Hastaların çoğu kadın olup siklofosfamid ve doksorubisin gibi kemoterapötik ajanlar kadınlardaki kanserlerde sık kullanılmaktadır. Bu kemoterapötik ajanlar germ hücrelerine belirgin toksik etkisi olduğu yapılan çalışmalarda gösterilmiştir. Vitamin D hem anti-proliferatif etkiyle hem de anjiyogenezi inhibe ederek hücrelerin apoptozisini engeller. Doksorubisin ve siklofosfamid'in over dokusunda oluşturduğu hasar üzerine vitamin Din etkisi belli değildir. Bu çalışmada doksorubisin ve siklofosfamid uygulanmasinin siçan over dokusunda ve fonksiyonlarında meydana getirdiği değişiklikler üzerine vitamin D'in etkilerinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Çalışmaya otuzaltı adet dişi rat alındı. Deney hayvanları her grupta 6 hayvan olacak şekilde rastgele, 6 gruba; Kontrol grubu, Doksorubisin grubu, Doksorubisin + Vitamin D grubu, Siklofosfamid grubu, Siklofosfamid + Vitamin D grubu, Vitamin D grubu ayrıldı. Deney sonunda tüm gruplardaki ratlar anestezi altında dekapite edildi. Over dokuları hızla çıkarılıp histopatlojık kesitler alınıp over rezervi incelendi. Bulgular: Kontrol grubu ve Vitamin D grubunda over rezervi iyi bulunurken diğer gruplarda overrezervinde azalma izlendi. En fazla azalmada siklofosfamid grubunda izlendi. Sonuç: Sonuç olarak, bu çalışmada Vitamin D ek dozunun uygulamasının, siklofosfamid ve doksorubisinin sitotoksik etkisini önleyemediğini görüldü
